Herpes simplex virus (HSV), mainly, HSV type 2, is the microbe which causes genital herpes (GH). An increasing prevalence of GH has a profound effect on the health of human beings in the world. GH caused by HSV type 1 increase gradually due to the changes of sexual behavior. Over the recent two decades, the emergence of more and more drug-resistant HSV strains has been associated with the widespread and frequent use of antiviral drug for treatment, suppressive therapy and prophylaxis of HSV infection, maturating of organ transplantation, and the increasing of HIV infection.Herpes simplex virus type 2 resistance to antiviral drug has been described primarily in the immunocompromised patients.Aciclovir{9-[(2-hydroxyethoxy)methyl)guanine} is the representative of nucleoside analogues, the most common antiherpes drug. It has been used primarily in the prevention and treatment of HSV infections. Despite controversy some authors considered that long-term use of antivirals would induce resistance due to survival stress on virus. Few investigation on ACV resistance were made in China. The susceptibilities of HSV-2 clinical strains isolated in Nanjing to ACV and VCV, and isolated ACV-resistant strains from reference strains and clinical strains by plaque purification were investigated.Chapter 1: Isolate HSV strains from clinical specimens and investigation on HSV infective activity link with different clinical phase.Objective: To isolate the HSV strains from patients' specimens and evaluate the infectious ability of patients with GH in different clinical phases, and to study HSV's cytopathic effects (CPE) in Vero cell lines.Methods: HSV strains were isolated in 50 samples from patients with GH,and typed with monoclonal antibodies by immunofluorescence assay.
