The metabolite profiles from rat hepatic fibrosis, nude mouse model of human pancreatic cancer and rat chronic pancreatitis were investigated using high-resolution magic-angle spinning ~1H nuclear magnetic resonance (HR-MAS NMR) and principal component analysis (PCA) on intact tissue.In study of the metabolite characters of hepatic fibrosis, we found elevated level of lipids, depleted levels of glucose, glycogen, taurine and inositol, and decreased levels of choline (Cho) and glycerophosphocholine (GPC) compared with that of phosphocholine (PC). There was a dramatic tendency observed for the level of lipids with the aggravated stages of hepatic fibrosis, which ascended greatly at first and descended a little bit at the last stage. However, no evident changes of other metabolites in different hepatic fibrosis stages were observed.In study of the nude mice of human pancreatic cancer, we found elevated levels of Ile/Leu, valine, lactate, alanine, Glx, choline, taurine and glucide, and decreased levels of GPC and an unknown peak (53.66) compared with that of PC. Compared the metabolite characters of tissue before treatment with three different Gy-treatment stages (10, 20, 30 Gray), we found few changes in the spectra, while the stronger the Gy-treatment is, the smaller the tumor is. But there seemed some regular changes in our PCA results, due to the contents of taurine and choline-contained metabolite (CCM).In study of the rat chronic pancreatitis, we found elevated levels of GPC, and decreased levels of TMAO&Bet, Glx, alanine, glycine, and an unknown peak (53.88) compared with that of PC. There are some varied metabolite characters owing to species differences between nude and rat mice, such as CCM, taurine, glycine, acetic, and so on. So it was difficult to compare pancreatic cancer with chronic pancreatitis at present.
