Objective To investigate proliferation inhibiting effect of shikonin on bladder cancer T24 Cells and its mechanism.Methods The bladder T24 cells were treated with shikonin in 25 μmol/L, 50μmol/L and 75 μmol/L for 12 hour, 24 hour or 48 hour. Cell Proliferation inhibition was determined with MTT.COX-2 mRNA exression level was assessed by reverse transcriptase polymerase chain reaction(RT-PCR), and exprssion of COX-2 protein was evaluated by western blot after the bladder T24 cells were treated with shikonin in 25 μmol/L, 50 μmol/L and 75 μmol/L for 24 hour or 48 hour.. At the same time, it was set up a control group (0μ mol/L) and a celecoxib group (50μ mol/L).Results 50 μmol/L celecoxib could inhibit proliferation of T24 cell , decreased expression of COX-2 mRNA and protein. Shikonin could significantly inhibit proliferation of T24 cell at concentrations from 25 to 75 μ mol/L. There were great statistical signification between varies medicated group and control group (P<0.05) ,and there also were great statistical signification between medicated group each other(P<0.05). Cells Proliferation inhibition rate increased in a dose and time-dependent manner and expression of COX-2 mRNA and protein was also decreased by various concentrations of shikonin in a dose and time -dependent manner.Conclusion Shikonin could significantly inhibit proliferation of T24 cell, COX-2 maybe involved in it.
