OBJECTIVETo study the effect of neural stem cells transplantation to the spinal cord after injuryMETHODSWe isolate and cultivated neural stem cells from the embryonic rat brain, then we confirm their potential of proliferation and differentiation that they were neural stem cells. We built spinal cord injury model in rats according to the a weight-drop device NYU impactor; Their score were no more than 4 by BBB locomotor rating scale, 36 healthy Sprague-Dawley rats were randomly divided into two groups: control group and NSCs group, each of them are eighteen. Culture medium was injected into spinal cord after injury in group A. And NSCs were transplanted into spinal cord after injury in group B on the 9th day after spinal cord injury. The function of spinal cord was evaluated by BBB locomotor rating scale at lweek, 2weeks, 4weeks, 6weeks after transplantation. At2weeks, 6weeks got the material and made slice of frozen section and immunohistochemistry, which would be used to observe the existence of NSCs and their differentiation in the spinal cord.RESULTSAll of them get a certain recovery by BBB locomotor rating scale, but the recovery is far from the normal. The rats of group B is got the better recovery than the other group's(p<0. 01) . The slices show that there are NSCs which were transplanted in the spinal cord, and the neurons in group B are much more than the other group (p<0.01).CONCLUSIONThe neural stem cells from the embryonic rat brain can be cultivated well in vitro and they do not differentiate easily, but the cultivation...
