Objective: The effect of the energy element media on the activity and active rate in human spermMethods: The semen samples were obtained from health volunteer aged 25-40 years old,with 3-7days abstinence. The whole semen was collected into sterile cup by masturbation. All the semen accepted for experiment met the normal criterion of World Health Organization.Laboratory Manual for the Examination of Human Semen and Semen-Cervical Mucus Interaction.3rd ed,and the antibody from semen plasm and blood serum were negative. Human Tube Fluid (HTF)was used as control .The ingredients related to energy in HTF were changed to observed the effect on the dynamic parameters of sperm.The ingredients of the culture media were as follows:①Experiment 1: The study group included subgroup 1 containing fructose(fructose 25 mg instead of glucose) and subgroup 2 without glucose.②Experiment 2: glycin 1.0mM, glutamine 1.0mM and taurine 0.1mM were added to HTF.③Experiment 3: pyruvate was replaced by n-Octanoic acid 0.329mM and n-Hexanoic acid 0.38mM.④Experiment 4: ADP 2.5mM and ATP 2.5mM were added to HTF respectively.⑤Experiment 5: L-Carnitine 2.5mM and Pentoxifylline 2.5mM were added to HTF respectively. Each semen sample was divided into 2 or 3 aliquots and then were treated with study group media and control group media, respectively .Data were analysed by SPSS 10.0.Results: In Experiment 1.the vitality and motility of sperm cultured for 48 hours and motility cultured for 72 hours showed statistically significant difference between study group and control group(P <0.05). Our data suggest that the fructose group was better than HTF , and HTF better than the none suger group. The fructose group and the HTF group are together better than the none suger group.
