The Experimented Study For Inhibitory Effects Of Ginsenoside Rg3 On Endometriosis By Peritoneal Injection

Objective To study the inhibitory effects of Ginsenoside Rg3 on Endometriosis by peritoneal injection and explore the mechanism, the author observed ginsenoside Rg3's influence on endometriosis rats model ectopic endometrium grafts volume ,morphology and angiogenesis. Methods Endometriosis models were established by autotransplanting endometium in mature female Wistar rats.Successful endomtriosis rats were divided control group, ginsenoside Rg3 5mg/kg,ginsenoside Rg3 10mg/kg group, gestrione group and ovariectomised group with 2 weeks, 4 weeks, 6 weeks and 8 weeks all 20 groups.Ectopic endometrium grafts volume and histomorphology were observed,the expression of vescular endothelial growth factor(VEGF) and thrombospodin-1(TSP-1) were observed using immunohistochemistry(SP technique) and indirect fluorescent method. Microvessel density(MVD) was determinded by immunostaining for CD31.And the vascular permeability of ectopic endomerium grafts were investigated by the evaluation of Evans blue leakage.Results (1) No significant difference was found in the volumes of grafts of each groups before therapy(P>0.05) and the appearance of the grafts are almost same. (2) No significant difference was found in the volumes of grafts of each groups and the appearance of the grafts are similar between the second and third operathion of control group rats. (3) The volumes of grafts of ginsenosid Rg3 10mg/kg treated rats are reduced compared with the control rats'and the simultaneous ginsenosid Rg3 5mg/kg treated rats',while no difference with the simultaneous ginsenoside groups.In Rg3 10mg/kg groups , the volume of grafts of the rats treated for 4 weeks are reduced than those treated for 2 weeks while no significant difference compared with those treated for 6 weeks and 8 weeks. (4) The expression of VEGF, TSP-1 and MVD of ectopic tissue of each control group were no significant difference(P>0.05). (5) the expression of MVD of the animals treated by ginsenosid Rg3 10mg/kg were singnificantly lower than those treated by ginsenosid Rg3 5mg/kg and gestrione after the same time of therapy, higher than those of ovariectomised groups. the expression of MVD of the rats treated for 4 weeks are reduced than those treated for 2 weeks while no significant difference compared with those treated for 6 weeks and 8 weeks. (6) The expression of TSP-1 of grafts in the rat treated by ginsenoside Rg3 10mg/kg 8ws is singnificantly higher than other rats except for ovariectomised rats for 8ws.And there was singnificantly linear negative correlation between expression of TSP-1 and MVD(r=-0.585,P<0.001).
