Backgrounds Fructus Xanthii is the mature fruit with involucre of Xanthium sibiricum Patr., corapositae plant. It is gathered when the fruit is mature in autumn and dried, removed away foreign matter such as stalk and leaf. Its nature and flavour is warm, pungent and bitter, toxicity , belonging to the lung meridian. Its function includes dissolving wind, dehumidity and smoothing orifices . Its indication is headache caused by wind-cold, nose running by rhinorrhea with turbid discharge , pruritus by rubella, contracture by arthralgia spasm. It is used to treat allergic rhinitis and other hypersensitive diseases clinically, indicating Fructus Xanthii has the function of anti-anaphylaxis reaction and a certain immunosuppression effect. The research group previous study and research shows, the traditional decoction of Fructus Xanthii has immunosuppression function.Objective To study immunosuppression activity beginning with the traditional decoction of Fructus Xanthii, then, find active site by the approach of system solvent extraction method and immunopharmacology experiments.Methods and results1 The experiment of the maximum tolerance dose (MTD) of orally takingmedication to miceThe experimental result indicated the maximum tolerance dose (MTD) of FructusXahthii orally taking medication to mice was 240g/kg. d, 1300 multiple much as the adult dosage clinically. Under the dosage, there was no obvious toxic action and adverse reaction.2 The influence of the decoction of Fructus Xanthii on I-hypersensitivity reaction— passive cutaneous anaphylaxis (PCA) of ratsMethods To study the inhibition effect of the decoction of Fructus Xanthii on I-hypersensitivity reaction, with the experiment of PCA.Results Measuring 0D of blue spot ,1:5 multiple diluting antiserum , compared with control group, the low dosage group of Fructus Xanthii has remarkable significance (P<0. 05), while the high dosage group of Fructus Xanthii , tripterygiura glycosides group and prednisone acetate group have very remarkable significance (P<0. 01);Measuring OD of blue spot , 1:10 multiple diluting antiserum , compared with control group, the low dosage group of Fructus Xanthii has remarkable significance (P<0. 05), while the high dosage group of Fructus Xanthii , tripterygium glycosides group and prednisone acetate group have very remarkable significance (P<0. 01). With the blue spot OD of two different multiple diluting antiserum , compared with positive control group , including tripterygium glycosides group and prednisone acetate group, the high dosage group of Fructus Xanthii has no remarkable significance (P>0. 05).Conclusion The decoction of Fructus Xanthii can obviousy suppress passive cutaneous anaphylaxis (PCA) of rats, indicating Fructus Xanthii has obvious inhibition on I-hypersensitivity reaction.3 System solvent extraction method separating immunosuppression active site of Fructus XanthiiMethods To separate the decoction of Fructus Xanthii by the approach of system solvent extraction method, then study the immunosuppression activity of the extraction sites ,with the experiments of immune organs weight of mice .elimination carbon from plasma, delayed type hypersensitivity (DTH) and hemolysin generative test.Results Compared with control group , the extraction sites groups ofFructus Xanthii have no remarkable significance (P>0. 05) on the immune organs weight of mice, detected by spleen index and thymus index;Compared with control group , ether extracts group has remarkable significance (P <0. 05) on the corrected clearance index a , elimination carbon from mice plasma;Compared with control group , ether extracts group has remarkable significance (P<0. 05) .while ethyl acetate (EAc) extracts group has very remarkable significance (P<0. 01) on the delayed type hypersensitivity (DTH) of mice;Compared with control group, the extraction sites groups of Fructus Xanthii have no remarkable significance (P>0.05) on mice hemolysin generative.Conclusion Fructus Xanthii has no obvious immunotoxicity on the immune organs including spleen and thymus. Ether extracts and ethyl acetate (EAc) extracts have the immunosuppression on the mononuclear macrophage immune system and specific cellular immunity of mice. So, ether extracts and ethyl acetate (EAc) extracts are the immunosuppression active sites of Fructus Xanthii.4 Study chemical constituent of active sitesStudy chemical constituent of ether extracts and ethyl acetate (EAc) extracts, by the approach of thin-layer chromatography (TLC),the results indicates , low polarity position includes Succinic Acid.Conclusion Fructus Xanthii has a indeed immunosuppression function and its immunotoxicity is tiny;Its low polarity position is the immunosuppression active site;The low polarity position includes Succinic Acid, and Succinic Acid maybe is one of the active constituents.
