Objective:To investigate the immune function of sentinel lymph node (SLN) and non-sentinel lymph node (NSLN) in breast cancer.Method: 30 patients with breast lumps were diagnosed as invading ductule breast cancer by pathological examination via sonography guide core needle biopsy before operation. Radioactive substance and blue dye were injected to identify the sentinel lymph nodes. According to the patient's individual condition modified radical mastectomy or breast-conserving surgery was chosen and all patients underwent axillary lymph node dissection.One sentinel lymph node and one non-sentinel lymph node were taken out to study for immune function in every patient. With every lymph node specimen, half of it was sent to have a pathologic examination, another half of it was prepared in unicellular float liquid and flow cytometry was used to analyze the immune function. Using two kinds of flow cytometry methods CD45/SSC-Gating and FSC/SSC- Gating to study the immune function of lymph nodes, and the results were compared each other. The better method was chosen to study the immune function of lymph nodes. Statistical analysis was performed using SPSS10.0.Result: The SLN were all successfully identified in 30 patients, the ratio of success was 100%. 16 SLN were found to have metastases in 30 SLN. 13 NSLN were found to have metastases in 30 NSLN. Every specimen was prepared in unicellular float liquid as thickness 4-6*10~6/L. After the flow cytometry analysis, the TL% of FSC/SSC-Gating was 79.42±5.02, TL% of CD45/SSC-Gating was 97.66±4.60(P<0.05). The CD45/SSC-Gating was better than the FSC/SSC-...
