Objective: To explore the changes of Bax and Bcl-2 in a rat model of acute myocardial infarction(AMI).Methods: Sixty male and female Sprague-Dawley rats were used in this study. Left anterior descending branch of coronary artery ligation produced extensive AMI in 35 of 55 animals which survived the initial surgery. The 35 surviving animals were divided into 7 time points,with 5 rats for each duration of infarction. The remaining five animals undergoing thoracotomy but not coronary ligation served as controls.Apoptotic index, the expression of Bax and Bcl-2 protein in myocardium were analyzed semiquantitatively by TUNEL and immunohistochemical assay 1,2,4,6,8,10,12 hours after coronary ligation.Results: (1)The distribution of apoptosis: apoptosis was mainly distributed at the area surroundings in the infarction.That is to say, apoptosis was in regions between infarction and non- infarction. The apoptotic index of apoptotic Cardiomyocytes in infarct area began to increase at 1h group and achieved the peak at 4h and then dropped gradually. (2)The permillage of the positive Cardiomyocytes of the expression of Bax protein began to increase at 1h group and achieved the peak at 4h and then dropped gradually at 12h. (3) The permillage of the positive Cardiomyocytes of the expression of Bcl-2 protein began to increase at 1h group and then dropped gradually ,achieved the least at 4h , increased to the normal gradually at 8h and higher than the normal at 12h.
