Effects of arecoline on contraction of antral circular muscle strips in guinea pigs.Recently some researches on monomers of Chinese herbs indicate that they can modulate gastrointestinal motility. All of these monomers are natural chemical compounds that are extracted and purified from Chinese herbs. The experiments of these monomers and their ramifications will promote their exploitations and clinical applications, and offer more choices for treating the gastric dyskinesis. So we select arecoline, a monomer of Chinese herb (betel nuts, a Chinese medicine named binglang) to study how arecoline has influence on contraction of antral circular muscle strips and its possible mechanism involved in guinea pigs with the method of perfusing isolated gastric muscle strips. The experiment results were as follows:1. Arecoline (10-7mol/L,10-6mol/L) increased the mean contractile amplitude of antral circular muscle strips. Arecoline (10-6mol/L) increased the resting tension of antral circular muscle strips.2. Cholinergic M3 antagonist 4-DAMP abolished arecoline-related response,but cholinergic M2 antagonist gallamine didn't affect the exciting action .This experiment results showed that arecoline can promote gastric motility of the isolated antral circular muscle strips in guinea pigs. The exciting action of arecoline was mediated via cholinergic M3 receptor, but not cholinergic M2 receptor.â…¡. Effects of radish seeds on contraction of antral circular muscle strips in guinea pigs.The gastric dyskinesis is frequent pathologic reaction of many chronic diseases of gastrointestinal tract and the complication of some diseases. It is characterized by decreasing gastrointestinal motiiity and retarding gastric emptying. The clinically used western medcines such as metoclopramide, domperidone, cisapride and alosetron usually have conspicuous effects of promoting gastric motility, but the side effects of Western medcines should be valued at the same time. The natural resources of Chinese herbs are abundant and side effectsare much less, so the experiments of Chinese herbs on gastrointestinal motility are prevalent. Recently some researches on radish seeds indicate that they can modulate gastrointestinal motility. The exciting action of radish seeds is mediated via cholinergic M receptor, but hasn't gone into M receptor subtype. All of these experiments focused on small intestine and ileum.No study or report about stomach has been published, so we studied how the water extracts of radish seeds affect the contraction of antral circular muscle strips and its possible mechanism involved in guinea pigs with the method of perfusing isolated gastric muscle strips. The experiment results were as follows:l.The water extracts of radish seeds with the concentration of 30%, 100% and 200% dose-dependently increased the mean contractile amplitude of antral circular muscle strips. The water extracts of radish seeds with the concentration of 100% and 200% dose-dependently increased the contractile frequency of antral circular muscle strips. 2.Cholinergic M3 antagonist 4-DAMP abolished the exciting action of antral circular muscle strips caused by radish seeds partly,but cholinergic M2 antagonist gallamine didn't affect the exciting action.This experiment results showed that radish seeds could promote gastric motility of the isolated antral circular muscle strips in guinea pigs.
