LDL's oxidate product is oxLDL. The research findings show that the increase of oxLDL is an important reason of the As formation, and is closely related to the occurrence rate and mortality rate of ASCVD (Arteriosclerosis cardiovascular disease).At present, As and relative disease is the first killer which lead to death not only in our country but also in western developed countries. oxLDL is the special component in As focus, and its strength is in proportion to the degree of pathological changes. As a diagnosis index, serum oxLDL is better than other general indexs. Through measuring the level of serum oxLDL, As can be assistant diagnosed in the forepart. Whether healthy persons or patients with high blood lipoprotein, It has important significance in many sides to detect serum oxLDL clinicly, such as diagnosis, the judgement of treat effect and early-warning and so on. Four cell strains which can steadily excrete McAbs(monoclonal antibodies) were established in this study, named F3, B7, G12, B4. The filtration, preparation, purification, identify of the McAbs and work partnership that are the foundation of the clinical trial were finishied. The F3 was used as a coating antibody, and HRP (Horse Radish Peroxidase) ladeled B7 as a signal antibody and a double antibody sandwish ELISA procedure was investigated. The optimal conditions and the involved reagents are detemined. Finally a DAS-ELISA detection kit was basically developed, and a reasonable result obtained. The primary clinical trial proved that the sensitivity and specificity of the kit was better.At the mean time The DAS-ELISA detection kit is the necessary foundation of the study of golden colloid kit, and one inventive patent was applied based on this study and the patents acceptance number is 2005102003946.
