The function of human body was very complicated. The urinary system took great responsibitities for discharging wastes. The cystic micturition was one important link that urinary system was responsible for important physiological function. We observed the contractile activity of isolated detrusor strips in Guinea pigs and explored the mechanism of Chinese traditional medicine and monomer for contractile activity of isolated detrusor strips in Guinea pigs.Emodin (1, 3, 8-trihydroxy-6methy lanthraquinone, FM:C15H10O5, FW:270.23), was widely present in plants and insects, especially in rhubarb. Some researches suggested rhubarb could dose-dependently increase resting tension, mean contractile amplitude and frequency in bladder detrusor strips. Because of the multiplicity of the ingredient of rhubarb, it was difficulty in both clinical application and study. Thus we studied the effects of emodin on contractile activity of isolated detrusor strips and deeply explored whether emodin had the same action with rhubarb and its mechanism. At the same time, we explored the new uses of Chinese traditional medicine. The radix astragali and folium sennae were broadly applied in clinical treatment. We also studied the effects of Radix Astragali and Folium Sennae on cotractile activity of isolated detrusor strips in Guinea pigs and their mechanism of action.Guinea pigs of the either sex, weighing 350-450g, were killed by stunning , then their urinary bladders were rapidly removed and placed in 37℃ Krebs solution. The ventral of the bladder was isolated from the trigon and the mucosa was removed. This tissue was cut into 2mm-wide and 6mm-long strips along the longitudinal direction. One end was suspended in a tissue chamber (95 % O2 and 5 % CO2;37 ℃) and the other in an external isometric force transducer. Motility of detrusor muscle strips was recorded on ink-writing recorders. After 1h equilibration under lg-load tension, various drugs were separately added into the tissue chamber. In the experiment to accumulate emodin, Radix Astragali and Folium Sennae per 2min. antagonists were applied in the experiment, including: L-type calcium channel blocker Verapamil(10"7mol/L);a -adrenergic receptor antagonist Phentolamine (10"6 mol/L);Ni receptor antagonist Hexamethonium (10"6 mol/L);M2-muscarinic cholinergic receptor antagonist Gallamine (10"6 mol/L);M3-muscarinic cholinergic receptor antagonist 4-DAMP(10"6 mol/L).All results were expressed as mean ± SD. The results of tension and mean contractile amplitude were demonstrated by percentage of the control contractile response. The frequency was calculated as the cycle of contractile waves per minute. Statistical comparisons were made using one-way ANOVA with the computer statistical package SPSS 12.0 (for windows). Differences with P<0.05 were considered statistically significant. Additionally, correlation coefficient (r) was calculated by statistical package SPSS 12.0 (for windows). The main results were as follows:Effects of emodin and antagonists on guinea pigs bladder detrusor strips in vitro:1. Effects of emodin on guinea pigs bladder detrusor strips in vitro:Administration of emodin dose-dependently increased resting tension (r—0.954, PO.01), mean contractile amplitude (r=0.944, PO.01) and frequency (r=0.980, P<0.01)in bladder detrueor strips.2. Effects of antagonists and emodin on guinea pigs bladder detrusor strips in vitro: (l).Verapamil partly inhibited the action of increased tension and mean contractle amplitude by emodin.(2). Phentolamine partly inhibited the action of increased tension and frequency byemodin.(3) Hexamethonium couldn't inhibit the contraction responses induced by emodin.(4).Gallamine partly inhibited the action of increased tension and mean contractleamplitude by emodin.(5).4-DAMP partly inhibited the contraction responses induced by emodin.The results suggest that the contraction responses induced by emodin may be via L-type calcium channel, M2-muscarinic cholinergic receptor, M3-muscarinic cholinergic receptor and a -adrenergic receptor.Effects of Radix Astragali and antagonists on guinea pigs bladder detrusor strips in vitro:1. Effects of antagonists and Radix Astragali on guinea pigs bladder detrusor strips in vitro:(l).Verapamil partly inhibited the action of increased tension by Radix Astragali.(2).Phentolamine partly inhibited the action of increased tension and frequency byRadix Astragali.(3).Atropine partly inhibited the contraction responses induced by Radix Astragali.(4).Hexamethonium couldn't inhibit the contraction responses induced by RadixAstragali.The results suggest that the contraction responses induced by Radix Astragali may be via L-type calcium channel, M-muscarinic cholinergic receptorand a -adrenergic receptor.Effects of Folium Sennae and antagonists on guinea pigs bladder detrusor strips in vitro:1. Effects of Folium Sennae on guinea pigs bladder detrusor strips in vitro: Administration of Folium Sennae dose-dependently increased resting tension (r= 0. 966, p<0.0\), mean contractile amplitude (r=0. 981, p<0.0l), and could increase frequency in bladder detrueor strips (PO.01, PO.001).2. Effects of antagonists and Folium Sennae on guinea pigs bladder detrusor strips in vitro:(1). 4-DAMP partly inhibited the contraction responses induced by Folium Sennae (2). Verapamil, Phentolamine and gallamine couldn't inhibit the contraction responses induced by Folium Sennae.The results suggest that the contraction responses induced by Folium Sennae maybe via M3-muscarinic cholinergic receptor.
