Gout and chronic nephropathy are higher morbidity disease. Hyperuricemia is the main causes of gout. Nephros-fibrosis is the end of development of chronic nephropathy, so decreasing blood uric acid and inhibiting development of nephros-fibrosis are the effective way of prevention and curing on the two diseases. Although synthetic medicine are often used, but take synthetic medicine for long time will bring people severe adverse reaction. So it have clinical meaning to select natural substances for curing the two diseases. The purpose of our study is selecting active substances extracted from natural plants, marine animal and microorganism and studying its mechanism on curing gout and inhibiting renal interstitial fibrosis in rats. Method: Through detecting the generation of uric acid and the NBT reduction induced by superoxide anions to measured the activity of these substances on xanthine oxidase. Mouse hyperuricemia mode were established by using hypoxanthine and the uncase inhibitor potassium oxonate. The acute gout arthritis model was established by injecting rats' ankle with monosodium urate solution. The renal interstitial fibrosis in rats was caused by unilateral ureteral obstruction(UUO). The inhibiting effects of rosmarinic acid, acteoside and renierol on xanthine oxdiase was observed in vitro and in vivo. And the effects of rosmarinic acid on gout arthritis in vivo and renal interstitial fibrosis in rats was also observed. The results show that rosmarinic acid, acteoside and renierol are activer in all samples. Their IC50 on xanthine oxidase are 1.85μg ? ml-1, 12.25μg ? ml-1 and 56μg ? ml-1 respectively. Rosmarinic acid, acteoside and renierol could reduce the level of uric acid in hyperuricemia mouse established by using hypoxanthine and the uricase inhibitor potassium oxonate. Rosmarinic acid also can lessen inflammation of gout arthritis established by injecting monosodium urate solution, and its effect perhaps relates to decreasing the expression of TNF- a in synovial fluid of treated rats. Rosmarinic acid can prevent renal interstitial fibrosis process in UUO rats and its effect is equivalent to ACEI . It's efforts maybe relates tothe decreasing of TGF- P ,.
