Object: To discuss the rational supporting individualized titration of immunosuppressive therapy. Method: ATP-bioluminescence assay (ATP-CVA) was used to determine the immune response of CD4~+ cells by ATP syntheses which stimulated by phytohemagglutinin(PHA-L). In this study, fifty normal healthy volunteers and ten recipients of liver transplants(pre and post-transplantation 1 , 2, 4 week) were assayed. Result: There was a linear relationship between the log ATP and the log luminescence in a range of 10~0-10~4ng/ml .Y=0.75X+2.37 (correlation coefficient r =0.99484) .The liver transplant recipients had the significant lower cell mediated immunity(CMI) function than normal healthy volunteers (p<0.05). There was a sharp decrease in immune function of liver recipients in the first week post-transplant, then gradually on the rise. Conclusion: Maybe using this assay to measure the immune function of liver recipients and monitoring the drug levels in blood can achieve a better rational supporting individualized titration of immunosuppressive therapy.
