The amount of bacteria in water is an important index which reflects the degree of the bacterial contamination directly.Now most water utilities and drinking water guidelines still rely solely on time-requiring heterotrophic plate counts(HPC)under the condition for the 24h and 37℃constant temperature culture.So the technology of fast and accurate monitoring of microbiological parameters in water will be the trend in the future.This paper concerned with the research of the practical application of ATP-bioluminescence.Through determining the plate counts and ATP-bioluminescence intensity of different OD600of laboratory stains,we find the equation of two methods.Then determine the water samples with more bacteria and the saled water with less bacteria.Improve the limit determination of the ATP-Bioluminescence for to be convenient and fast reaching the demand of hygienic test.First we find the good linear relationship between plate counts and ATP-bioluminescence intensity through determine the different OD600of stains.We get the result about the ATP contents of E.coli 1.38×10-18mol/cfu and the detection limit of OD6005.0×10-7.The ATP contents of S.aureus 1.25×10-18mol/cfu and the detection limit of OD6002.0×10-5.On the basis of the known stains of laboratory,we can calculate the bacteria level by the equation of ATP-bioluminescence intensity and plate counts in order to quickly determine the amount of the bacteria.In this paper,the technology was applied in different types of water bodies (freshwater body,market saled mineral water and water fountains),and systematic analysed the influence degree of various factors including plankton,free ATP and so on and propose measures to remove the interference to optimize the ATP bioluminescence assay to provide a scientific basis to monitor the bacteria counts in the water.The results showed that:two methods showed large discrepancy in monitoring the bacteria amount of bacteria in Liwa Creek,Yinchu Lake and Suzhou Creek.The bacterial content of bottled mineral water failed to detect out for the sensitivity of reagents and equipment limitations.The result of mineral water added with a certain concentration of bacteria show that the two methods coincide.Therefore the ATP bioluminescence assay has an excellent application value in the laboratory stains,but for complex samples containing a variety of micro-organisms,the method needs further exploration and research.
