Objectives Since mitochondrial OXPNOS plays an important role in the glucose-induced insulin secretion in pancreatic beta cells.the relationship between mitochondrial gene and the development of diabetes mellitus has been widely attended. Our purpose is to study the prevalence of mitochondrial DNA 5178A/C polymorphism and 8296A/G mutation in type 2 Diabetes mellitus patients and healthy controls among Chinese Han population of HuBei Province and mitochondrial DNA 5178A/C polymorphism association with serum lipid levels in type 2 diabetics.Methods PCR and RELP techniques were used to examine 174 patients with type 2 diabetes(diabetic group) and 207 healthy people (control group) to analyse the mitochondrial gene and detect the serum lipid levels in patients with type 2 diabetes mellitus.Results The frequency of 5178A in diabetic group and control group were 35% and 32% respectively and in the 5178C were 65% and 38% respectively. There were no significance difference between the two groups, for the frequencies of 5178A/C polymorphism genotypes The serum levels of LDL-C, HDL-C, TG, apoA, apoB, Lp(a) had no significance difference in the 5178A and 5178C but the serum TC level was significantly lower in the 5178A than 5178C. The mitochondrial DNA mutation at position 8296 was not found in any of the patients and controls.Conclusions the mtDNA 5178A/C polymorphism may not be related to the etiology of type 2 diabetes. However,5178A seems to have an antiatherogenic effect, at least in type 2 diabetic. The prevalence of the mitochondrial DNA8296A/C mutation was exactly very low and it may not a major cause of type 2 diabetic patients in China.
