Objective: The alteration of expression Gaq/11 in the liver were studied during acute respiratory distress syndrome(ARDS) and propofol pre-treatment to explore the reference for the therapy of the disease. Methods: To establish an ARDS model in rats by intravenous oleic acid.48 male and healthy wistar rats were randomly divided into control group given equal volume normal saline(C, n=8), pre-treatment group received propofol at 80mg. Kg~-1.h~-1through left internal jugular vein forhalf hour before given OA (0.2 ml.Kg~-1)(P, n=8)and oleic acid(OA, n=32) groups.According to time limit, OA groups were divided into 4 groups: OA30min, OA60min, OA90min,OA120min after injected OA at 0.2ml.Kg~-1 though tail vein.Blood gas, plasma LDH MDA ALT GOT ALP and liver homogenate LDH activity and MDA content were assessed during the experimental period and the concentration of Gaq/11 were measured by Western blot. Results: Compared with control group, PaO2 of OA groups and pre-treat-ment group obviously decreased(P<0.01), but PaO2 of pre-treatment group was higher than that of OA groups(P<0.01). pH of OA30min group was higher than that of control group(P<0.05), furthermore OA 90min and OA 120min groups were markedly reduced (P<0.01). pH also reduced in Pre-treatment group, compared with control group (P<0.01), but was higher than that in OA 90min and OA120min groups(P<0.01). compared with control group(C group), MAP of OA groups were lower and in decending tendency with time passing (P<0.05 or P<0.01). Serum and hepatic homogenate LDH activity and MDA content of OA groups were increasing and at OA 120min,they were thehighest(P<0.01). The measurement of hepatic function showed that ALT and GOT content of OA groups were more higher than those of C group, and there were significant difference between OA 90min 120min groups and C group(P<0.01). ALP content of OA groups have no difference with C group. The concentration of Gaq/11 of OA 90min, 120min were respectively increased to(134.34±19.14)%, (151.31± 23.67)% and it was in ascending tendency with time passing. MAP of P group was the lowest among all of groups. There were no difference of index of LDH in serum between P group and C group. But P group was a little higher than C group, but lower than OA 90min, 120min in plasma in liver.LDH activity and MDA content of P group were lower than OA 90min, 120min groups (P<0.05orP<0.01) and there was no remarkable difference between P groups and C groups.ALT and GOT content of P group were higher than those of C groups(P<0.05), but compare with OA 90min 120min groups, they were lower(P<0.05). In comparision of C group, The concentration of Gaq/11 of P group had no change, while it was lower than that of OA 90min 120min groups. Conclusions: Up regulation of the expression of Gaq/11 may play a role in ARDS liver injury. Pre-treatment with propofol can decrease the expression of Gaq/11 in the liver and relieve the injury of the liver during ARDS.
