Objective: Observe the change of Gq/11 protein concentration in the heart and kedney of rats during acute respiratory distress syndrome (ARDS). and propofol pre-treatmeat effect to explore the pathophysiological mechanism of ARDS. Methods: 48 male Wistar rats were randomly divided into experimental groups (OA groups, n=32) given oleic acid from tail vein (OA 0.2ml/kg), pre-treatment group (P group, n=8) given 1% propofol (80mg/kg/h) from left internal jugular vein for 30 minutes before given the equal dosage OA, and observed for 90 minutes, and control group (C group, n=8) given normal saline (0.2ml/kg). OA groups included 4 groups: OA30min, OA60min, OA90min, and OA120min. Blood gas indexes (pH, PaO2, PaCO2), cardiac function indexes (MAP, LVSP, ± LVdp/dtmax), renal function indexes (BUN, Cr) and enzyme indexes (MDA, LDH, CK, ACE) of plasma and two organs were measured. Gq/11 protein concentration in two organs was examined by Western blot. Results: PaO2 of OA groups and P group decreased (P<0.01), and PaO2 of P group was higher than that of OA 90min group (P<0.01). MAP of OA groups and P group obviously reduced (P<0.05 or 0.01), and MAP of P group was lower than that of OA 90min group (P<0.01). With time passing, cardiac function indexes (LVSP, ±LVdp/dtmax) of OA groups gradually decreased. The indexes of OA 90min, 120min groups and P group were lower than those of C group (P<0.05 or 0.01), and there was no difference between OA 90min groupand P group (P>0.05). BUN and Cr in the plasma of OA groups (except OA30min group) and P group notably increased (i><0.01), and the indexes of P group were higher than those of OA 90min group (P<0.05 or 0.01). MDA content and LDH activity in the plasma, kedney and heart of OA groups (except OA30min group) significantly increased (P<0.01). MDA content in the plasma and two organs, and LDH activity in two organs of P group were higher than those of C group (P<0.05 or 0.01), but lower than those of OA 90min group (P<0.05 or 0.01). CK activity in the plasma and heart of OA groups (except OA30min group) and P group notably increased (P<0.01), but the index of P group was lower than that of OA 90min group (P0.05). Compared with C group, Gq/11 protein concentration in the heart of OA groups (except OA30min group) and P group respectively increased to (124.72±24.05)%, (140.61 ±22.41)%, (150.84±24.49)%, (131.94±16.00)% (PO.05 or 0.01), and that in the kedney increased to (123.98+25.74)%, (149.34 ± 20.04)%, (154.69 + 24.44)%, (124.68±19.38)% (PO.05 or 0.01). Compared with OA 90min group, Gq/11 protein concentration of P group in the heart had no change (P>0.05), and that in the kedney reduced (PO.05). Conclusions: Up regulation of Gq/11 protein concentration in the heart and kedney may play a role in two organs injury during ARDS. Pre-treatment with propofol can relieve the harm, which has some relation with reducing Gq/11 protein concentration.
