| Objective: Suo Suo grape ( Vitis amurensis Rupr) , the traditional herb in Xinjiang province in China has abundant resource. The former scholars studied the chemical constituent of fruit and obtained one active component, which is a fatty compound: Oleanolic acid. So on the base of the former research, we have studied the chemical constituents of the fruit of suo suo grape. At the sane time, suo suo grape can be used as the medicine, which heve got very extensive appliance in the hospital of Xinjiang Uygur Hospital. We have studied the granules of suo suo grape with Xinjiang Uygur Hospital .We have changed the oral liquid of suo suo grape and studied the suo suo grape Granules for corring easily. Methods: At first, three kinds of fatty compounds were obtained from acetone ultrasonic extraction and two water solvable compounds have been isolated from water extraction. Then we adopt the way of alcohol and aqua abstraction ,because the active constituents of suo suo grape have not only fatty compounds but also water solvable compourds. The alcohol extraction of suo suo grape was screened by means of Optimal experiment design. Basing on the content of extractum and total tretepernoids and total flavonpids, we study four factors of the extracting process including extracting time, the amount of adding alcohol, and concentration of alcohol.To study three factors of the extracting process including extracting time, the amount of adding water, boiling time, taking polysaccharide content and water extracting content as variableparameters. And uniform-design is used to optimize the extracting process conditions. And then appropriate excipient were selected on the base of dissolution rate and to establish the quality control stand for granules. Results: At present, we have finished appraising the structures of five compounds by chemical reaction and spectroscopic means such as UV, 'H-NMR, 13C-NMR, 2D-NMR and identified them as B -sitosterol; oleanolic acidi 3 -sitosterol-glncopyranose glycoside. Rutin and Isoqui-noline ferulic acid-3-oxide-glncopyranose glycoside. Except oleanolic acid, the other compounds were isolated from suo suo grape for the first time. The alcohol eatracting process: 9 times amount of alcohol as much added and extracted 3 times, lhour each time, the water extracting process: 14 times amound of water as much as herbs was added and extracted 2 times ,1 hour earch time. Excipient formula consisted of extruction powder and lactose (1:2.5, w/w). Methods: The study shows the average recorery of oleanolic acid was 101.2% and RSD is 1.12%. The method is sensitive, accurate and reliable. Conclusion: The studies not only supplied and perfected the research data of the chemical constituents from the fruit of suo suo grape but also established firm foundation for further development of suo suo grape. At the same time the experimental results provide the data of quality standards of the Suo Suo Grape Granules. |
