Balima (Sambucus chinensis Lindl.) is a herb medicine which belongs to Sambucus,Caprifoliaceae. It is also called Lu Ying, Jie Gucao, Choucao, Ba Lengma, Qi Yejin, Qi Yema.Its attending functions are blood stasis, expelling wind and rolling, sweating, diuretic. It isusually used for bruises, rheumatism, dislocations, nephritis, edema, beriberi, edema, urticatia.LU YING PIAN (WS3-B-0294-90), LU YING KE LI (WS-10990(ZD-0990)-2002), LUYING CHONG JI (WS3-B-0295-90), LU YING TANG JIANG (WS3-B-0296) are sold onthe market. Their function attends for Shuganjianpi, blood circulation, swelling diuretic foracute viral hepatitis. Balima distributed in Shaanxi, Gansu, Jiangsu, Anhui, Zhejiang, Jiangxi,Fujian, Henan, Hubei, Hunan, Guangdong, Guangxi, Sichuan, Guizhou, Yunnan, Tibet andother provinces. It is also found in Japan.The study focuses on standards developments of Balima, such as identification anddetermination methods, the index components of the extraction process. The results are asfollows:1. After researching on all the information about the herb, it is finally determined as asource of whole plant from Sambucus, Caprifoliaceae;2. The most typical characteristics of Balima shape was inverted-cup fleshy flowers. Formedicinal powder identification, the characteristics included wood fiber, brown slug material,spongy tissue, single-threaded catheter, textured catheter and a small amount of bordered pitscatheter, the crystallization of calcium oxalate.UA and OA are the standard material whendeveloped TLC method for Balima medicine.3. Build high-performance liquid chromatography (HPLC) method for determination ofUrsolic acid (UA) and oleanolic acid (OA). A good linear relationship were showed when theinjection amount range of2.16-10.8μg(UA),1.68-8.4μg(OA), respectively(RUA2=0.9999;ROA2=1).The average UA content was0.875mg·g-1,the minimum UA content0.650mg·g-1,while the highest value was1.312mg·g-1.For OA, the average value was0.279mg·g-1,the lowest value to be0.195mg·g-1,the highest to be0.407mg·g-1. The average recovery ofUA and OA come to99.18%and100.13%respectively. The RSDs were2.95%(UA, n=5) and2.45%(OA, n=5). The method is simple, accurate with good reproducibility. It can beused in determination of UA and OA content in Balima.4. Determination results suggested quality control standards set: the moisture contentshould be below12.0%; the total ash content should be less than11.0%, acid-insoluble ashcontent of less than7.0%; ethanol-soluble extract content of not less than6.0%; ursolic acidand oleanolic acid content of the total amount of not less than0.1%.5. Ursolic acid (UA) and oleanolic acid (OA) were selected as the quality evaluation ofpharmacological compounds according to their medicinal principle. The HPLC method ofthe simultaneous determination of two components was established. Different power, volumefraction of methanol, liquid ratio, extraction time, temperature, the extraction of ursolic acidand oleanolic acid process were optimized through single-factor test and Box-Behnkencentral composite response surface method. The optimized results were as follows: under theconditions of90%methanol,50°C for the extraction temperature, liquid feed ratio of25:1,extracting for30min ursolic acid rate was up to1.518mg g-1(RSD3.45%), oleanolic acidyield of0.501mg g-1,(RSD2.31%).
