Objective To observe the therapeutical effect of Sheng Qi Jiang Tang Ke Li on diabetic rats with insulin resistance and infer possible mechanism and target of action according to the resuts of observation. Methods The experiment was carried out at the animal experimental lab of Affiliated Hospital of Qingdao Medical College from 2004-10 to 2005-07. â‘ 90 healthy male Wistar rats were selected and randomly divided into 6 groups as Sheng Qi Jiang Tang Ke Li low, middle, high dose group, rosiglitazone(Avandia) group, type 2 diabetes mellitus model group and normal group. The diabetic rats model with insulin resistance was established by high fat diet and injection of streptozotocin(30mg/Kg). â‘¡The normal group and type 2 diabetes mellitus model group were given distalled water 1mL/100g each day. The concentration of solution of Sheng Qi Jiang Tang Ke Li was equal to that of crud drug 2.5g/mL and the dose was coverted directly through human-rat unit body weight and 5 times as low dose,10 times as middle dose,20 times as high dose. While the dose of rosiglitazone group was converted through the surface area of human and rat and rats in this group were given 10% suspension of rosiglitazone by intragastric administration. When the rats' plasma glucose(GLU) were decreased to normal level or there was significant different comparing with the original were defined as effective and put to death. After three months administration, all the rats would be put to death whether effective or not. Weight, blood urea nitrogen(BUN), creatinine(CRE), total cholesterin(TC), triglyceride(TG), insulin(Ins) of different stage of the rats were also measured. â‘¢ELISA of serum angiotonin II (Ang II )and endothelin-1(ET-1);immunohistochemical determination of ET-1 and protein kinase C(PKC) in aorta and type I acceptor of Ang II(AT1) in heart;semi-determination of mRNA expression of ET-1 in aorta. â‘£Difference between measurement data were compared through analysis of variance and that between groups with Q test. Results â‘ After one month of high fat diet, the weight and insulin were increased significantly. After injection of streptozotocin, blood glucose was increased significantly. After administration, the difference of blood glucose, blood urea nitrogen, creatinine, total cholesterin, triglyceride, insulin and weight of the experimental groups of different stages were significant. Blood glcose of Sheng Qi Jiang Tang Ke Limiddle, high dose group and rosiglitazone group were decreased significantly while insulin of the three groups were significantly higher than type 2 diabetes mellitus model group, but all less than normal control group. There was no significant difference between Sheng Qi Jiang Tang Ke Li middle, high group and rosiglitazone group in the reduction of glucose and insulin. Total cholesterina and triglyceride in all the experimental groups also decreased remarkably. ?ELISA:The level of Ang II and ET-1 were both significantly reduced in rosiglitazone group and Sheng Qi Jiang Tang Ke Li middle and high dose group comparing with the diabetic model group. ?Immunohistochemical determination:The expression of ET-1,PKC and ATI were all remarkably reduced in rosiglitazone group and Sheng Qi Jiang Tang Ke Li high dose group comparing with the diabetic model group. ? Semi-determination of mRNA expression of ET-1 in aorta:The expression of ET-1 mRNA was significantly reduced in rosiglitazone group and Sheng Qi Jiang Tang Ke Li high dose group comparing with the diabetic model group. Conclusion Sheng Qi Jiang Tang Ke Li has the effect of anti-diabetes and sensitization of insulin and the effect assumes dose dependent. It can protect and improve the function of the endothelium of aorta and myocardium through reducing the experession of ET-1, Ang II (ATI) and PKC.
