The mortality of the benign prostate hypertrophy ( BPH) and the prostate cancer (PCa) are increased. Since there was rare symptoms during early stage of Pca, when they really come, the local and far metastasis had already appeared. The major cause of sufferers' death is the development to metastasis phase of tumor. Therefore the research of tumor aggregation and metastasis has become a hotspot.Expression of Thymosin β is little in normal tissues, while its expression in tumor is clearly higher than that of its original tissue, but the catholicity of its expression and the significance to tumors early diagnosis and prognosis are needed to be study. At present both inland and overseas are devoid of research of Thymosin β15 (Tβ15) in prostate cancer. So I assessed the presence of Tβ15 in prostate cancer and analysed the relationships between Tβ15 expression and prostate cancer' s differentiation and metastasis, in order to explore the significance of metastasis and prognosis.Materials and methods1. MaterialsTotal 61 prostate cancer tissues were clearly diagnosed,49 tissues came from the First Affiliated Hospital of Chinese Medicine University between 1999 and 2004;12 tissues came from the Central Hospital of Shenyang Medical College between 2000 and 2005;another 19 BPH and 2 normal prostate gland tissues came from the 202 hospital of PLA;fixed in 10% formaldehyde and embed-ded in paraffin.2. Clinical and pathologic dataAll 61 prostate cancer patients are male, from 42 to 84 years old,the average age is 67. 81,among them 4 are younger than 50 years old;5 tumor volume are less than 1 cm;The high,middle and low differentiation cases are 34,19 and 8 respectively;8 tissues had lymphatic metastasis.3. Major agents' sourceThe primary antibody anti - human thymosin beta IgY antibody were provided by E. Lvaniov in Greece Immunopeptide Chemistry Lab "NCSR DEMOKRI-OS". The HRP marked rabbit anti -chicken second antibody were bought from Shanghai Casarray Company. The Ultro - sensitive S - P kit and DAB agent kit were bought from Fuzhou Maxin Biological Company.4. Methods4.1 Immunohistochemistry for T(315Immunohistochemical staining was performed using an Ultro sensitive S — P kit according to the instructions provided by the manufacture. Color was developed using DAB/H2O solution. Negative comparison processed with PBS instead of the primary antibody.4. 2 Judge of resultsPositive result refers to the appearance of brown yellow in cytoplasm. After the areas of tumor tissue had been chosen randomly under low power, 1000 cells within tumors were counted in all together 10 fields at 200 magnification. Cases defined as positive were regarded by both staining intensity and the percentage of immunoreactive tumor cells.4. 3 Statistical analysisStatistical Package For Social Science ( SPSS ) statistical software ( version 11.5) was applied for data analysis. Chi square test was used to compare the clinicopathological characteristics with the gene' s expression. A P value less than 0. 05 was considered significant.Results1. The Tp15expression in PCa, BPH and normal prostate gland.In PC a, Tpi5 expressed diffusely in the cytoplasm, and with a high expression rate of 70.49% (43/61) , there has no T|315expression in the mesenchyma and paraneoplastic tissue,and no expression in normal prostate gland and BPH either.2. The relationship between the high expression of Tp15and the clinical pathologic factors.There are statistical significances between the Tpi5 expression and the age, tumor volume, high to low differentiation, and lymphatic metastasis of PCa.Conclusions1. The T(315 is highly expressed in PCa, but has no expression in BPH and normal prostate tissue, which indicates that Tj515may become a biomarker of PCa judgement;2. There are statistical significances between the Tf$15 high expression and age, tumor volume and lymph node metastasis;3. There is statistical significances between the Tpi5 high expression and high to low differentiation.
