Objectives: Photodynamic therapy is a photosensitive reaction with oxygen, which has a concomitant biology effect and be used in clinical therapy. Because of its selective characteristic, the method shows its importance step by step in the therapy of tumour and becomes a routine measure. At the same time, PDT was used in blood purification and inactive the contagious viruses in it. We hope we can find a effective method to inactive virus through our research.Merhods: 1. To research the effect of inactiving HSV-1 by photodynamic therapy via HpD in vitro. Avoid the effect on normal cells by PDT, irradiate HSV-1 cultured in vitro with two doses of intensity of illumination(2J/cm~2,6J/cm2) and certain concentration of photosensitive, culture cells in incubator of 37℃% 5%CO2 for 5 days , observe CPE and detect the survival ratio of cell with MTT. 2. Discuss the effect on hepatitis B virus in blood by photodynamic therapy. The transgenic mice transferred three gene segments were used in this experiment, irradiate through the vein of the frail by fibre after injection, the laser power is 5mW, the time is 30min.Observe the action of mice during the experiment, after 2 hours, compare the content of HBsAg in blood by enzyme linked immunosorbent assay (ELISA). 3. Primary research about the security. Irradiate the blood via jugular with three doses of HpD injected through caudal vein, check blood routine and other biochemical index. Analyze the effect of PDT.Result: In the first experiment, there was statistical difference in the content of virus between the irradiated and the virus control which indicate that photodynamic therapy can inactive HSV-1. About PDT inactive hepatitis B virus in blood, the experiment data shows that the content of HBsAg declines in various degree, photodynamic therapy can control the HBsAg in blood , and HbsAg mostly in the lipid bilayer of virus capsid which is very important to the virus infection. In the experiment of research on the security of PDT, the data fluctuate to a certain extent within the normal scope, we can think photodynamic therapy through irradiating the blood has no damaging effect under certain condition, but we should also verify its influence on others antigens and agents.Conclusion: Under certain concentration of HpD and intensity of illumination photodynamic therapy can inactive virus effectively in vitro. In the effect on certainnormal blood index by photodynamic therapy with irradiation on blood., we should choose the most appropriate concentration of photosensitive, on a base of validity, we should find the minimal damage to the normal agents.
