Objective: To study the methods to culture the bladder smooth muscle cells in vitro and evaluate the biocompatibility of polyhydroxybutyrate(PHB) combined with bladder smooth muscle cells.Methods: Rabbit bladder smooth muscle cells were separated and cultured by two methods, and then the cells were cultured with PHB in vitro. Cells were examined by phase contrast microscopy and SEM to assess cell growth and morphology. Cell adhesion and cell proliferation were detected with MTT assay.Results: Bladder smooth muscle cells growth well by both cultured methods; Bladder smooth muscle cells adhere well to PHB with the adherence rate 83.6%. PHB has no effects to the survival and proliferation of bladder smooth muscle cells. No obvious morphological changes were observed by phase contrast microscopy and SEM.Conclusions: All these indicated that Bladder smooth muscle cells can be cultured in vitro by these two methods. PHB has a good biocompatibility with bladder smooth muscle cells, PHB has a potential to be used as scaffold in urinary tissue engineering.
