Alcohol is the major component of alcoholic beverage and is abused generally in the world. Over drinking will hurt central nerval system, heart and brain vein, digestive system, peripheral nerval twig and it can also result in spirit obstacle, heart failure, poisonous encephalopathy, liver disease (alcoholic liver, fatty liver, hepatocirrhosis), extensive neuritis, etc. Over drinking may induce abnormal behavior, accident even crime, for example, it can cause traffic accident, strangle and trouble.Blood alcohol concentration (BAC), which is primary gist dealing with affairs, needs to be detected under many circumstance. After drinking, BAC will change from time to time. Some scholars once tried to confer BAC at a certain time or drinking dose using alcohol pharmacokinetics. But alcohol pharmacokinetics will be influenced by multiple factors such as drinking fashion, kind of alcoholic beverage, food, enzyme and so on. Some scholars have been paid attention to it but little results reported. We want to establish an alcohol detection method, which is suitable for many circumstance. We will detect volunteers' BAC at different intervals using the method and then investigate how the kind of alcoholic beverage, drinking time and consumer influence alcohol pharmacokinetics. The results can provide groundwork for further researching about alcohol pharmacokinetics.In daily work we may take over all kinds of materials, in which there arenot only alcohol but also acetaldehyde, methanol, formaldehyde and n-propyl. These substance may influence detection for alcohol, some may be interested in by us. To set up an applicable widely determination method for alcohol, we investigated conditions of headspace, gas chromatogram and so on, then method for alcohol in biological samples were developed as follows: headspace incubative temperature 60, transmission line temperature 125, loop temperature 125, incubative time 10 minutes, interval time 10 minutes, gas chromatogram column temperature 48, injector temperature 220, FID detector temperature 250, carrier gas N2 flow rate lmL/min, H2 flow rate 30mL/min, air flow rate 300mL/min, split ratio 20:1, detection time 8 minutes. Qualitative gist is reserved time. Quantitative method is internal standard method, using isopropyl as internal standard. Quantitative base is the ratio of alcohol peak area and isopropyl peak area.This method was evaluated by linearity, recovery, within-day and between-day precision. The method is accurate and precise, total detection time is no longer than 18 minutes. The method proposed can be used for analysis alcohol in biological samples and suitable for decomposed materials. The method is suitable for many circumstance and methanol, formaldehyde, acetaldehyde can also be detected using this method.The pharmacokinetics of alcohol was investigated in 9 healthy men after oral administration of 50g/man. Blood samples were collected at different intervals after alcohol administration, and the concentration-time curves are acquired. The result showed that the pharmacokinetics of alcohol could be fitted to one-compartment model in man and the weight is 1. The result also showed that in the same drinking time, drinking beer comparing with drinking distilled alcohol, absorption is faster, Cmax is higher, Tmax is earlier and clearance isfaster. The dose and the kind of alcoholic beverage are both same, drinking in short time comparing with drinking in longer time, absorption is faster, Cmax is higher, Tmax is earlier and clearance is faster, hi the same drinking time, the dose and the kind of alcoholic beverage are both same, regular consumer comparing with irregular consumer, absorption is slower, Cmax is lower. Alcohol pharmacokinetics is different among individual, even the same person in different conditions. All these hint us that if we want to confer BAC in a certain time or drinking dose we should consider the influence on alcohol pharmacokinetics.The method for alcohol in biological samples was established in this paper. Influencing factors on alcohol pharmacokinetics were studied. The results provide groundwork for further work about alcohol pharmacokinetics and offer valuable gist for dealing with affairs involving alcohol.
