The Effect Of Estrogen And Progesterone On Mouse Endometrial Leukemia Inhibitory Factor (LIF) Espression | | Posted on:2006-02-13 | Degree:Master | Type:Thesis | | Country:China | Candidate:H Q Pang | Full Text:PDF | | GTID:2144360155973405 | Subject:Obstetrics and gynecology | | Abstract/Summary: | PDF Full Text Request | | Objective To investegate the effect of estrogen and progesterone on mouse endometrial LIF espression ,which is involved in the receptivity establishment.Methods Ovariectomized mice were randomly divided into three control groups and seven treatment groups .Estrogen in two different concentrations (oestradial-17 β 100ng /d and 1000 ng/d ) was given to two treatment groups respectively for one week.The two concentrations estrogen combined with progesterone (progesterone 2mg/d or 500mg/d) regimens were given to the other four treatment groups respectively for one week.The control groups was subdivided into ovariectomized group,shamp group and positive group (day 4 pregnancy) .Medicine was administered subcutaneously.The expression of LIF and LIFmRNA in mouse endometrium is evaluated by streptoavidin peroxidase conjugated (SP) immunohistochemistry and in situ hybridization. Results1. The LIFmRNA and LIF protein mainly is localized to the uterine luminaland glandular epithelia.The expression of LIF is weak in uterine stroma.2. The exprssion of LIF in the endometrial cells of the ovarectomized mouse is negative .there is significant diferrence between ovarectomized group and shamp group (P<0.05) .3. The exprssion of LIF in the mouse endometrium treated with physiological concentrations of estrogen is higher than that of supraphysiological estrogen group (P<0.05) .4. The exprssion of LIF in the endometrium of the physiological level of estrogen combined with progesterone group is higher than that of supraphysiological estrogen combined with progesterone group (.P <0.05) . there is no difference between hysiological estrogen combined with physiological concentrations of progesterone group and positive group .5. The expression of LIFmRNA is parelling with that of LIF protein.
