ObjectiveIn this study, we detected the expression and localization of Peroxisome proliferator-activated receptor Y (PPAR Y ) in the term pregnancy placentas and fetal membranes before active term human parturition and after active term human parturition. We discussed the relationship between the expression of PPAR Y in pregnancy tissue and the onset of labor and elucidated its role of onset of labor.MethodsSpecimens were collected from women with term singleton pregnancies normal vaginal deliveries (NVD group=20) and selective cesarean section before labor(CS group=20) to prepare for RT-PCR and immunohistochemistry analysis . Immunohistochemistry was used to detect the protein expression of PPAR Y in placentas and fetal membranes including before onset of labor and after onset of labor, Immunohistochemistry was used to localize each of these tissues , which stain intensity and positive cell percentage were half-qualitatively assessed by the computer with picture analyzing software; RT-PCR was used to detect the mRNA expression of PPAR Y in placentas and fetal membranes including before onset of labor and after onset of labor. Gel imaging analysis device was used to analyze the results of the agarose gel electrophoresis and to take photographs. The results were indicated by optical density of the PPAR Y / 3 -actin.ResultsTwo groups all exhibited protein expression of PPAR Y in fetal membranes and placentas by immunohistochemical analysis . Immunostaining was prominent innuclei of amnion cell and chorion cells,nuclei of cytotrophoblast cells and syncytiotrophoblast cells, nuclei of endothelial cells in villous blood vessels .Howere, immunostaining was absent in the mesenchymal core of the chorionic villi. In be for onset of labor group exhibited stronger staining for PPAR-y in fetal membranes and placentas than in the after onset of labor group (PO.05). RT-PCR analysis revealed the mRNA expression of PPAR-y in normal term fetal membrances and placentas. PPAR-y mRNA expression of fetal membrances and placentas in CS group was significantly higher than in NVD group(P<0.05). The results were same as Immunohistochemistry showed.â–  ConclusionPPART is highly expressed in term fetal membranes and placentas, PPART levels are reduced once active labor commences.The changes with labor suggest that regulation of PPAR7 expression and function may play a roles in the mechanisms of maintaining pregnancy or initiating labor. This research reveal the mechanisms of onset of labour and provide a new method for prevention and therapy of preterm labour.
