The parathyroid hormone (PTH) is one of important regulatory peptides secreted by parathyroid glands, and plays an important role in development and differentiation of bone. PTH includes 84 animo acids. Recent studies showed that the administration of low dose PTH intermitently could improve bone formation, so it was considered as the most promising drug in the treatment of osteoporosis in clinic. In addition, it was reported that the PTH 1-34 fragment has the similar biofuction as the intact PTH 1-84 in bone formation.In this study, we employed the recombinant DNA technology to produce recombinant human parathyroid hormone 1-34 fragment.The human parathyroid human gene was inserted into the expression vector-pThioHisA, then the construct was transformed into E.coli BL21(DE3) or DHSastrains. We express the recombinant PTH 1-34 fragment in E.coli. The expressed product was confirmed as PTH 1-34 by Western-blot, and its bioactivity was detacted by its effects on improving the cAMP concentration of 3T3 cells.
