Objective:Investigate the therapeutic effects of small molecular peptide CMS010.26 and its therapeutic mechanisms.Methods:1. Investigate CMS010.26 effect on T lymphocyte transform in vitro and in vivo with MTT method. Investigate CMS010.26 effect on antibody formation with hemolysin method.2. Kidney cortex homogenate was injected ip to establish rat active Heymann nephritis model; Purified antigen Fx1A was injected on footpad to establish rat active Heymann nephritis model.3. Heymann nephritis model were established. All the animals were detected 24 hours urinary protein, serum BUN, CR, serum albumin, total cholesterol, total triglyceride, weight and kidney index. Kidney pathological damage was studied.4. Culture rats primary mesangial cell. Study CMS010.26 effect on the mesangial cell proliferation and TNF-α level excreted by mesangial cell.Results'.1. CMS010.26 can inhibit T lymphocyte proliferation in vitro and in vivo, can inhibit antibody formation in vivo with 200μg/kg, 100μg/kg/d 和 50μg/kg/d dose.2. Compared the two methods of model establishment, we found the second method had a earlier and higher success rate. It existed significantly increased 24h urine protein, BUN, CR and serious pathological damages coincidence with MGN. So we selected the second method in the formal experiment.3. CMS010.26 was used to therapy rats Heymann nephritis. The urine protein level decreased significantly from the 8th to the 14th week. Serum creatinine, blood uria nitrogen, total cholesterol and total triglyceride decreased, serum albumin increased. All of the above results showed statistic significant...
