Objective: Human recombinant basic fibroblast growth factor (b-FGF) is a potent mitogen. b-FGF is both a powerful stimulator of angiogenesis and a regulator of cellular migration and proliferation. First.we succeed in culturing embryo chicken's tendon cell.Then we studied the effects of b-FGF on cell proliferation with MTT test. Materials and methods:1 The embryo chickens are cultured in a cubator.At day 19, the 10 embryo chicken is killed to get the tendons each time. Tenocytes are obtained from tendon segments after being processed by enzymes.Then the tendon cells are cultured with 20% foetal calf serum (FCS) culture medium .2 Tenocytes are cultured in 96-hole culture plates with different kinds of culture medium. The b-FGF and 20% foetal calf serum (FCS) are added to serum-free culture medium to firm 5 groups (lng/ml, 5ng/ml, 10 ng/ml ,50 ng/ml ,200ng/ml) whose FCS concentration is 2%. They are compared with other 2 no b-FGF groups whose FCS concentration is 2% and 5%. Then we studied the effects of b-FGF on cell proliferation with MTT test.3 The effects of b-FGF on cell proliferation is tested with Swiss-Gimmsa taint test.Results:1 The contour of tendon cell is like a fibroblast.lt grows very fast after it sticks to the wall.2 The proliferative rate of tendon cell suggest a basic difference in different concentration of foetal calf serum (FCS) culture medium.3 We get the conclusion through the MTT test after we culture the tendon cells with differern concentration of b-FGF FCS culture medium .(1) The second day reasults:The proliferative rate of tendon cell suggest a basic difference indifferent concentration of b-FGF.The b-FGF at lOng/ml was found to be the most effectiveconcentration.There are no difference among the concentration of b-FGF atlOng/ml ,50ng/ml, 200ng/ml.(2) The fourth day reasults:The proliferative rate of tendon cell suggest a basic difference indifferent concentration of b-FGF.The b-FGF at lOng/ml was found to be the most effectiveconcentration. The proliferative rate of tendon cell at that concentrationis the fastest.The proliferative rate of tendon cell get fast if tendon cells is culturedwith higher concentration of FCS.All these results are got through analysis of variance. Conclusion:1 The tendon cell has its own proliferative rule.The tendon cell proliferative rate get fast after it go down to the future generation.
