Objective To explore whether bone marrow stem cells from adult rat can be differentiated into hepatocyte-like cells by hepatocyte growth factor (HGF) and the time phase characteristics in the differentiation progress. Methods Adult rat bone marrow stem cells were treated with or without HGF, on days 0, 7, 14, 21. The morphologic characteristics of the cells were observed; The expression of AFP and CK18 protein were detected immunohistochemically by using anti-AFP and CK18 antibodies. Glycogen were tested by periodic acid-schiff (PAS); the albumin (ALB) and AFP mRNA were analyzed subsequently using reverse transcription polymerase chain reaction (RT-PCR). Results â‘  Freshly isolated adult rat bone marrow stem cells expressed AFP mRNA, AFP weakly, but didn't express ALB mRNA, CK18, and produce glycogen. In the group without HGF, AFP mRNA , ALB mRNA , AFP, CK18, glycogen could not be detected in every time phases. But in the HGF treated group, the AFP mRNA was positive at all times, however it tended to decrease after day 7; the expression of ALB mRNA, CK18 protein and glycogen -positive cells increased on the time -dependent manner.â‘¡ In the group without HGF, the diameter of the small circular cells which dyed deeper by hematoxylin-eosin didn't change in every time phases; But in the HGF theated group, the diameter of the small circular cells which dyed deeper by hematoxylin-eosin and the cells which expressed AFP, CK18 protein , glycogen increased in the differentiation progress. Conclusion â‘  Adult rat bone marrow stem cells could be induced into the hepatocyte-like cells by HGF in vitro. The expression of the markers of hepatocyte coincided with the development of liver cell. â‘¡ The morphologic characteristics of the hepatocyte stem cells from bone marrow is the small circular cells which dyed deeper by hematoxylin-eosin.
