Objective : To investigate the cilinical significance of expression of Prox-1 mRNA and VEGF-C mRNA in colorectal carcinoma. Method : Reverse transcription-polymerase chain raction was used to examine the expression of Prox-1,VEGF-C,VEGFR-3,LYVE-1mRNA in tumor tissues,tumor surrounding tissues and normal tissues in 25 colorectal carcinoma,and Immunohistochemical streptavidin-peroxidace was used to examine its Lymphatic vessel density. Result: The expression of Prox-1,VEGF-C,VEGFR-3 mRNA in tumor tissues and tumor surrounding tissues was significance higher than normal tissues(P<0.05).The expression of VEGF-C mRNA in tumor surrounding tissues and tumor tissues had significant difference in the group of Ducks A+B stage and Ducks C+D stage and the group of with lymph node metastasis and without lymph node metastasis (P<0.05). The expression of Prox-1 and VEGFR-3 in tumor surrounding tissues had significant difference in the group of Ducks A+B stage and Ducks C+D stage and the group of with lymph node metastasis and without lymph node metastasis (P<0.05). The lymphatic vessel density(LVD) in tumor and tumor surrounding tissues have had significant difference(P<0.05);the LVD in tumor had significant difference in the group of Ducks A+B stage and Ducks C+D stage and the group of with lymph node metastasis and without lymph node metastasis (P<0.05).The expression of VEGF-C and VEGFR-3 in surrounding tissues and tumor tissues had significant linear correlation(P<0.05),the coefficient correlation was 0.551 , 0.653.The expression of VEGFR-3 and Prox-1 mRNA in tumor surrounding tissues had linear correlation(P<0.05) with LVD,the coefficient correlation was 0.472,0.537(P<0.05). Conclusion: (1)The overexpression of Prox-1 induced the lymphatic endothelial cell differentiation.(2)The overexpression of VEGF-C VEGFR-3 may enhanced the generate of lymphatic vessels in the stroma of colorectal carcinoma, which lead to the cancer cells can easily invade the lymphatic vessels. (3)Prox-1,VEGFR-3 and LVD is anticipation factor, but is not a independent factor. (4)When used these factor to anticipated should choose the tumor borderline tissue.
