OBJECTIVE: The expression of the aromatase cytochrome P450 messenger ribonucleic acid(P450 arom mRNA) on hyperandrogenism and anovulation /oligoovulation was investigated in polycystic ovary-like syndrome rats ovaries. METHODS: The polycystic ovary-like syndrome model was established by subcutaneous injection of 1.25 mg testosterone propionate to Sprague -Dawley female rats at the age of 9 days. Rats were sacrificed around 100 days of age to determine circulating levels of testosterone(T),estradiol(E2),follicular stimulating hormone (FSH),luteinizing hormone (LH),fasting plasma glucose(FPG) ,fasting insulin(FINS) and ovarian P450 arom mRNA levels. RESULTS: polycystic ovary-like syndrome rats manifested with PCO, anovulation/oligoovulation, elevated body weight,hyperandrogenism and hyperinsulinism. E2 ,FSH,LH levels ,E2/T ratio and P450 arom mRNA expression were decreased, and FPG,FINS levels,LH/FSH ratio and Homa IR were elevated in ovary-like polycystic syndrome rats. Positive correlations between T and FINS, between E2/T and P450arom mRNA,and between FINS and P450 arom mRNA were observed. CONCLUSION(S): Our data suggest that T injection to immature female rats can induce PCO,anovulation/oligoovulation,hyperandrogenism and hyperinsulinism. P450arom mRNA expression were decreased in polycystic ovary-like syndrome rats ovaries. Moreover, the E2/T ratio as an index of aromatase activity for the diagnosis and therapy of PCOS.
