Objective: To evaluate the association with the C242T A640G polymorphism of the p22phox gene, an essential component of NAD(P)H oxidase in Chinese Han people with type 2 diabetes mellitus. Method: C242T and A640G polymorphism of the p22 phox gene was detected by polymerase chain reaction-restriction fragment-length polymorphism in 194 Chinese type 2 diabetic subjects and 105 health subjects. While we examined the association with this mutation and the patients' clinical characteristics such as fast blood glucose, fast blood insulin, TG, TC, HDL, LDL, body mass index, waist circumference, hip circumference, and HbAlc.RESULTS: The diabetic subjects with the TC+TT genotypes (20.62%vs3.81%) displayed a significantly higher than those of health subjects (p=0.002), the allele genotypes C and T had a great difference between the two groups (83.25%,16.75%vs97.14%,2.86% p=0.000). There were no differences between the AG+AA genotypes (12.89%vs12.38% p=0.562) or G and A allele genotypes (85.96%,14.04%vs87.62%,12.38% p=0.195) in the two groups. Logistic multiple regression analysis shows T242 allele, fasting blood glucose and HbA1c were independent risk factors of Type 2 diabetes.CONCLUSIONS:These results demonstrate that p22phox C242T gene polymorphism is associated with T2DM in Chinese Han people. P22phox subunit T allele mutation probablly is one of predisposing genes of Chinese Han T2DM.,and show that p22phox A640G gene...
