Objective: Through observing the influence of different Staphylococcus Epidermidis (SE) in different liquid culture to Dacron segments, which preincubated for 120 minutes at 37° with plasma or ones not do, on Bacterial Biofilm formation and the effects of antibodies against the purified ESS on bacterial biofilm formation. We can provide beneficial way for the purpose of the prophylaxis of Bacterial Centered Infection (BCI).Methods: One group was in normal liquid culture, the others was in culture with the concentration of Nacl and glucose to 2% and 3% respectively. Some Dacron segments were preincubated for 120 minutes at 37℃ with plasma and some were not do, then taking the experiment of bacterial biofilm formation to SE and to its adhension-positive, accumulaton-negative mutant M7. The density of Bacterial Biofilmassociated with Dacron segments were meatured by Confocal Laser Scanning Microscope (CLSM). At the same time, we use the Scanning Electron Microscope (SEM) to observe the configuration of BF on the surface of Dacron segments. Results:1. There is a obvios difference of density of BF on Dacron segments between RP62A and M7 (P<0. 05);2. Plasma-preincubated Dacron segments increase the density of BF obviousely compare with none-treated ones in RP62A group (P <0. 05);3. Antibodies against the purified ESS can reduce the thickness of BF and then blocking the formation of BF (P <0.05);4. Adjusted group of liquid culture have more density of BF than control group (P<0.05);5. BF formation summit phase was 24-30 hours surveyed from SEM and CLSM, during which ESS could be observed.Conclusions: BF formation on the surface of biomaterials was a dynamic process which summit . time was 24-30 hours. Both plasma-preincubated Dacron and adjust liquid culture can stimulate the bacteria to secrete ESS and promote the formation of BF. On the contrary , antibodies-added groups can decrease the density of BF remarkably and block the formation of BF. Our experiment indicated that ESS play a great role in BF formation on the surface of biomaterials and the blockage of the BF formation will give us further advance in treating BCI.
