| Halichondria rugosa belongs to the genus of Halichondria. family of Halichondrida. In recent years, many constituents with antibacterial, anticancer. antiviral activities have been isolated from the marine sponges of the halichondria. These active constituents (macrolides. terpenes. sterols. alkaloids) attracted the interesting of foreign chemists of natural products. There is not any report about the chemical constituents of the genus halicondria in our country. In order to discover marine natural products with new structures and bioactive activities, we have investigated the marine sponge halichondria rugosa (Ridley & Dendy). The halichondria rugosa distributed abundantly in South China Sea. especially in Hainan Province. HL-60 and BEL-7402 bioassay-guided fractionation led to the isolation of the active constituents. Fifteen compounds have been isolated from the ethanolic extract of Halichondria rugosa by repeatedly chromatography on normal and reversed-phase silica gel columns. Sephadex LH-20. and final purification by normal and reversed preparative HPLC By detailed analysis of the spectral data (IR. EIMS. ES1MS.~1HNMR. ~13CNMR. DEPT. COSY. NOESY. HMQC, HMBC) and chemical methods, nine of them were elucidated as following: 24. 25-dimethyl-3 β -hydroxy-5-chole-stene-2 β -sodium sulfate( I ). 5-cholestene-2. 3-oxide (II). 3-[(16-hydroxy-17 E)-tetracose-nyloxy]-l,2-propanediol (III). 24ξ. 25-dimethyl-5α-cholestane-2β.3α.6α-triyl trisodium sulfate(IV), 24ξ. 25-dimethyl-5-cholestene-2β.3α-diol( V ). uracil(VI). P-sitosterol(VII). stigmasterol(VIII) and batyl alcohol (IX). All the compounds were obtained from the halichondria rugosa for the first time. Compounds I . II. III are novel natural products. Some of them were found to show significant bioactivities in vitro against human tumor cell lines. Such as. the IC50 of compound I against the human tumor cells-BEL-7402. HT-29. SPC-A1. U-251 were 0.023. 0.012. 0.006. 0.007 umol/L. respectively. These can help people to recognize the constituents of halichondria rugosa and also apply to the science research of the sea. |
