Objective To explore the relationship between merlin and hippocampal sclerosis of temporal epilepsy by observing expression of merlin in cortex of temporal lobe and hippocampal CA1 region of the kindling model of epilepsy induced by corciaria lactone (CL) in rats.Methods 25 healthy male SD rats were divided into experimental group (n=20) and control group (n=5) randomly.The experimental group received 1ml/Kg CL intramuscular injection every three and a half days. The control group received the equal amount of normal saline solution. After two and a half months, the rats whose behaviors reach 3-5 grade (Racine scale) for 10 times during the course of that the rats were injected for two times continuously belong to kindled group , and the others belong to non-kindled group. EcoG of all groups rats were recorded. HE staining method and TUNEL technique were used to examine the morphology of brain and the apoptosis of neruons.To observe expression of merlin in neuron of cortex of temporal lobe and hippocampal CA1 region of all brain tissues fixed with Paraformaldehyde by means of immunohistochemistry and image analysis method.Results Neuronal apoptosis was observed significantly both in cortex of temporal lobe and hippocampal CA1 region of kindled group. The expressionof merlin in neuron of cortex of temporal lobe and hippocampal CA1 region of kindled group were more than that of the other two groups.(P<0.05), and the expression of merlin in the non-kindled group was not significant difference from that in the control group .(p>0.05).Conclusion The upregulation of merlin in neuron of cortex of temporal lobe and hippocampal CA1 region of the kindled rats may be involved in the process of neuronal apoptosis.
