| Objectives: Emodin (1, 3, 8-trihydroxy-6-methylanthraqui-none) is an active herbal component traditionally used in the treatment of various ailments in China. And it exerts potent therapeutic effects on acute pancreatitis, but the actual molecular mechanism of action is still not clear. Since acute pancreatitis is associated with systemic inflammatory response syndrome, we investigate whether emodin attenuate inflammatory response in the rat lung following severe acute pancreatitis.Methods: SAP model was induced by injection of 3% sodium taurocholic acid into the subcapsular of pancreas. Emodin was administered by injecting into the intestines. Plasm amylase of 3h, 6h and 12h with SAP was measured. Expressions of IL-1 β , IL-6 and IL-10 mRNA in lung were detected by semi-quantitative RT-PCR. Immunohistochemistry was used to assay the expression of ICE in the rat' s lung.Results: Emodin attenuated plasm amylase level , the pathological lesion score of lung, the mRNA expression ofpro-inflammatory cytokin IL—1 3 and IL-6, as well as the expression of ICE. Moreover, we demonstrated for the first time that emodin upregulated the mRNA expression of IL-10.Conclusion: Emodin exerts anti-inflammatory effects on the rat lung following severe acute pancreatitis by negatively modulating the mRNA expression of IL-1P IL-6 and positively modulating the mRNA expression of IL-10. |
