Objective: Lacrimal gland and salivary gland distribute only in head and neck, not like bone and ligament. When these glands are destroyed irreversibly by some diseases, it is unfeasible to regenerate them with the patient's own cells by means of engineering technique. Using heterogeneous acinar cells from the glands of other people or animals is an alternative, however it will arouse immunoreaction. Thus, scaffolds which can isolate cells from immunocyte and immunoglobulin of the host must be used. In the study, we make an initial investigation on the feasibility of polyethersulfone (PES) used as this kind of scaffold for artificial lacrimal glands.Methods: (1)Acinar epithelial cells from lacrimal glands of Sprague-Dawley rats (20-25 days of age) were cultured in vitro. Cells were examined with transmission electron microscope and detected a-smooth—actin, vimentincel and celll-asssociated SC by immunochemistry technique and immunofluorescence microscope , respectively. (2)The permeability of ascorbic acid, glucose, L-tryptophan and IgG was examined for hollow one-close-end tube made of PES. (3)PES membranes were seeded with the 2th passage of acinar epitherlial cells invitro, and examined with scanning electron microscopyResults: (DAcinar epithelial cells from lacrimal gland of Sprague-Dawley rats have been cultured in vitro. And the 2th and 3th passage of the cells in vitro could be used as seeding cells of artificial gland. (2)The one-close-end hollow PES tube can isolate cells from immunosystem of host like IgG, and allow nutritions like ascorbic acid, glucose and L-tryptophan pass through. (3)PES membranes can support the growth of acinar epithelial cells from lacrimal gland of SD rats. Conclusions: PES could be used as the scaffold of artificial lacrimal gland .
