| Objective: Mercury is a extremely special metal and the only liquid metal at normal temperature in the natural world,which is applied widespread in industry and agriculture. The health of human can be damaged because exposed to extremely low dose mercury.Mercury and it's all kinds of compounds can nearly reach everywhere of human bodies,which is in liver initially,then transferred to renal slowly,especially in proximal convoluted tubule.To renal,the target organ,the most important compound of merury is inorganic mercury such as mercuric chloride.The purpose of the research is to approach the related mechanism of the toxicity exerted to biology and cells by mercuric chloride as well as the antagonism and the mechanism of selenite and zinc, which antagonize the toxicity of mercuric chloride from the view of genotoxic effect of DNA damage induced by mercuric chloride on biology and vero cells, can provide experimental data for the future steps carried out to prevent the exposed population from the toxicity.Methods: The research is divided into two parts.The vero cells are studied in the first part.Firstly,we studied the proliferation toxicity of mercuric chloride to vero cells with the MTT assay and offered the bases in dosage designed in the latter parts; secondly,the genotoxicy of mercuric chloride on vero cells and the inhibition of Zinc and Selenite added together or respectively are studied with the SCGE(single cell gel electrophoresis) assay;the effect of apoptosis of mercuric chloride on vero cells are studied used Confocal Scanning Laser Microscope(CSLM).Crucians are studied in the second part of my research.The genotoxicity that mercuric chloride exerted to Crucians was studied used with the SCGE assay;at the same time ,we studied the changing of SOD and MDA in the three tissues of crucians exposed to low dosage of mercuric chloride.ConcIusion:The proliferation of vero cells exposed to low dosage of mercuric chloride in short time can be inhibited significantly and the genotoxicity induced by mercuricchloride can be detected significantly.Zinc and selenite used together or respectively can inhibit the effects induced by mercuric chloride to some extent,at the same time,we found that different combines of concentrations of Zinc and Selenite can attain different inhibitive results such as the concentration combine of middle dosage of Zinc and middle dosage of Selenite inhibited the toxicity of mercuric chloride effectively. Low dosage of mercuric chloride can lead vero cells to apoptosis and the rate of apoptosis increase with the rise of dose .We conclude that lipid peroxidation and free radicals participate in the process and mechanism of toxicity induced by mercuric chloride combined with other datas before.Genotoxicity and DNA damage can be detected in the body of crucian exposed to water populated by mercuric chloride together with the changing of activity of some enzymes exiting in the body of crucian. We conclude that the pollution of water can be detected easily using the assays mentioned above together with other physical and chemical assays in the future. |
