| Flos Datura is the dry flower of Datura metel L. which belongs to the family of Solanaceae. It has the long history to be used as traditional Chinese medicine for a long history and utilized widely to cure many diseases such as cough, asthma, convulsion, and psoriasis etc. for its strong and wide biological activities. There have been many reports about its chemical constituents, pharMacolo gical effects and clinical studies. In these years, Flos Datura preparations have been reported to have significant curative effects on psoriasis with many advantages for its high rate of remedy effects and low rate of relapse, however the chemical constituents against psoriasis have not been studied and found yet. In order to elucidate the effective constituents of Flos Datura and its action mechanism against psoriasis, and provide important scientific basis to discover and develop new drugs, the effective part and the constituents of Flos Datura were studied according to the previous studies against psoriasis on the basis of the results of pharmacodyn amics.In the course of our studies on chemical constituents in effective parts of treating psoriasis, 16compounds were isolated with silica gel , reversed-phase (ODS) , Sephadex LH-20 column chromatography and HPLC , of which the structures of 12 compounds were fully determined according to the chemical and physicochemical evidence (ID and 2D-NMR). 2 novel withanolide compounds were firstly reported and 7 compounds were firstly found in Datara metel L .Among them there are 5 withanolide compounds, 3 sesquiterpenoids compounds and 4 others.The structures of the two novel withanolide glycosides wereidentified as , DaturametelineB ( 5α,6β,15β,21-tetrahydroxy-l-(20R,22R) -oxowitha-24-enolide), Daturameteline C (5α,6β,12β,21 -tetrahydroxy-27-0-methyl- (20R.22R) -1- oxowitha-2,24-dieno...
