Objective: The aim of this study is to observe the influence of plasma substitutes ( HydroxyEthylStarch ) with different concentrations on IL-2, 10,12 and IFN- γ of normal persons, which will prove if HydroxyEthylStarch restrains normal persons immune system.Methods: 12 normal volunteers, each one supply venous blood 20ml in sterility, and blood is anticoagulated by heparin. In super clean bench, single nuclear cells are separated from the blood. After counting the cells is divided into five groups: 1640 culture medium negative controlling group, 10% plasma group, 10% HydroxyEthylStarch group, 20% HydroxyEthylStarch group and 40% HydroxyEthylStarch group. Then each group is arranged into negative controlling group, adding LPS group, adding PHA group. The cells are cultured in each culture medium for 48 hours. After centrifuge, the superior clear fluid is extracted from the culture medium. Enzymerlinked immunosorbent assay (ELISA) is used to detect the level of IL-2, IL-10, IL-12 and IFN- γ in superior clear fluid.Results: Comparing trial groups with controlling groups in the three conditions (negative control, adding LPS, adding PHA) shows: Although the level of cytokines in each group has a little difference, they haven't significant statistic discrepancy (p>0.05). In IL-2 and IFN- γ , there are not significant statistic discrepancy between negative controlling group and adding LPS group (p>0.05), but both have significant statistic discrepancy with adding PHA group (p<0.05).
