Objective : The gastrointestinal peristalsis is of great importance to the gastrointestinal tract and the physiology function. The gastrointestinal peristalsis not only transmits chyme, but promote the discharge of the bacteria and its outcome in the intestinal tract. It also has a usual immunity defense function to gastrointestinal tract. It is known that under the conditions of trauma, operation, infection, Gastric motor dysfunction usually occur. Its clinical symptoms are: anorexia, abdomen distention, nausea and vomiting. If not ameliorated, serious aftereffect may result. At present, gastric motility promotion treatment is the key to the release of gastric motor dysfunction. Huoxiang drugs contain rich volatile oil that can eliminate wetness, invigorating the spleen and the stomach. The research of pharmacology and the effect of medicine show that Huoxiang drugs and Huoxiang preparation have a usual function of protection to the intestinal mucosa dysfunction of various disease mechanism, promote olfaction, gestation and gastric mucosa, increase gastric mucosa blooding, excite intestinal peristalsis, quicken gastrointestinal advance, accelerate the transferring of epithelium cell and lipoprotein, and improve the activity of digestive enzyme and other functions. Attention: Such medicine and Huoxiang preparation can not only adjust, protect the structure and function of the intestinal barrier, they are also effective to the adjustment of the gastrointestinal movement function. In order to further investigate the function of gastric movement regulation , gastric motor dysfunction and the mechanism of Huoxiang drugs, the study employs Huoxiangzhengqi preparation as a representative, starting from the research of the clinical efficacy of Huoxiangzhengqi Soft Capsule to gastric motor dysfunction , to observe theinfluence of Huoxiangzhengqi Soft Capsule to the clinical symptoms and electrogastrograph(EGG) dominant frequency(DF) and direct power(DP) of the patients with gastric motor dysfunction before .Then the Wister rat gastric smooth muscle strips were used as the object of the research to observe the influence of HXZQ to the swing of the contract action and its function of adjustment to the gastric motility from the perspective of histology.Because the contract and relaxant action of gastric smooth muscle cell is the basis of the gastric movement, this study not only uses insolated single gastric smooth muscle cells of lower limb ischemic-reperfusion Wister rat as a model on the base of former experiment but also probes the signal transduction mechanism of the gastric motor dysfunction and the functional mechanism of the HXZQ under stress by observing the influence of length and form of gastric smooth muscle cells, NO level of gastric smooth muscle cells, and the effects of selective inhibitor of PKG KT5823 and PKA H-89 to smooth muscle cells length in view of the standard of cell molecular.Methods:1. To discuss the clinical efficacy of Huoxiangzhengqi Soft Capsule in the treatment of the patients with dysmotility-type functional dyspepsia(FD) by observing the clinical symptoms and EGG dominant frequency and direct power change before and after administration.2. To probe its functional mechanism of the adjustment to the gastric motility from the perspective of histology by observing the effects of HXZQ on the swing of the contract action gastric smooth muscle strips.3. Insolated single gastric smooth muscle cells model was induced from lower limb ischemic-reperfusion Wister rat, Groups with different HXZQ concentrations and reaction time were created, to investigate the proper cell concentration and reaction time by observing the contraction of smooth musclecells4. On the basis of the former experiments, observed the influence of the effects of selective inhibitor of PKG KT5823 and PKA H-89 to smooth muscle cells length and NO level of gastric smooth muscle cells, study the signal transduction mechanism of the relaxant action of the gastric smooth muscle cells of lower limb ischemic-reperfusion Wister rat and HXZQ functional mechanism.Results:1 The clinical symptom scores of the patients with dysmotility-type FD decreased after administration(P<0.01).2 Postprandial dominant frequency(DF) and direct power(DP) of gastric body, greater gastric curvature, gastric antra on EGG were lower than preprandial DF and DP before administration(p<0.05), they increased after the administration, Postprandial DF and DP after the administration were higher than that before administration(p<0.05).3 HXZQ obviously increased the contract swing of gastric smooth muscle strips of Wister rats and reinforce Smooth muscle contract ability (p<0.01) .4. Single gastric smooth muscle cell model was induced from lower limb ischemic-reperfusion Wister rat. the gastric smooth muscle cells of lower limb ischemic-reperfusion Wister rat were relaxant. The cells contraction was decreasing (p<0.01) .5. HXZQ reduced the NO content of model rats gastric smooth muscle cells (p<0.01) , increased the cells contraction (p<0.01) , The NO content decrease and cells contract were according to the drugs concentration.Conclusion:Huoxiang preparation have the following Clinical Efficacies: it can restore...
