| This paper adopted the supercritical CO2 extracting method to extract the active ingredient from Xanthochrous nilgheriensis Teng. In order to gain the best extracting technology of Xanthochrous nilgheriensis Teng, the influences of different factors on the acquisition were evaluated by orthogonal-designing method using L9(34) table. In the studies, we optimized the best method of making the original emulsion using the stabilized parameter Kg, the stable emulsion was acquired from the original emulsion with high pressure homogennization. The best prescription was screened by orthogonal- designing method using L9(34) table with the index sign of Zeta electric potential.The primary stable experiment was researhed, and the results showed that the emulsion had good physicochemical stability, and the new formulation can be tolerant at 121℃ for 30min.It was observed from the pharmacokinetics experiment in the rabbits that the pharmacokinetics parameter was remarkably changed when the acquisition from Xanthochrous nilgheriensis Teng was made into intravenous emulsion. Compared with the dissolving parts , the t1/2 of emulsion was significantly higher from 5.351h to 9.871h. We also studied the tissue distribution characteristics of the Xanthochrous nilgheriensis Teng intravenous emulsion, the result demonstrated that the emulsion distribution in rabbits had a definite direction to the lung organizes, and the quantitative evaluation parameter of TDDS was 17.26. |
