| Objective:To disscus the inhibiting effects of tetrandrine on haze after PRK in rabbits and explore it,s mechanism.In order to find out a new drug which can replace corticosteroid to treat haze after PRK. Methods:Sixty rabbits were randomly distributed into five groups:Tet group(0.05%,0.1%,0.2%),FML group,NS group.Haze was observed with Slit Lamp . Aqueous humor and coneal were obtained on the postoperative 1w,2w,1m,2m. The ultrastructure tessiue were observed with light microscope and transmission electron microscope.The change of collagen type Ⅲ , fibronection were measured by immunocytochemistry. The content of TGF-β1 in aqueous humor were observed with ELISA. Results:1 Observed with Slit Lamp after PRK surgery,haze in Tet group were relieved than FML group and NS group.2 Collagen type Ⅲ,fibronection on Tet group indexes were decreased(P<0.01).4 Collagen type Ⅲ,fibronection on FML group and NS group were no significant difference .3 The content of TGF-β1 in aqueous humor were increased after PRK.2 The content of TGF-β1 in aqueous humor on Tet group was decreased than FML group and NS group(P<0.01). Conclusion:1 Tetrandrine has obvious inhibitive effect on haze after PRK. 2 Tetrandrine may exert it,s inhibitive effect by decreasing ECM and releasing TGF-β1 .3 Tetrandrine has obvious inhibitive effect on corneal stroma fibroblast after PRK. 4 It shoud be further studied on the drug,s safe dosage scope and so on. |
