Objective: to investigate the changes of humoral factors and the regulation effects of shenfu injection in canine with cardiogenic shock after acute myocardial infarction and to elucidate its possible mechanism. Methods: twenty five hybridization canine were randomly divided into sham-operated group,model group, dopamine group, shenfu injection group and shenfu injection+dopamine group,According to Creamer method,the models of cardiogenic shock after acute myocardial infarction in canine were established when mean arterial pressure(MAP) was decreased to below seventy percent. The plasma NO,NOS iNOS levels were determined by colorimetric method, the activity of SOD were measured by xanthine hydrocarbonylation , The plasma MDA level was detected by TBA.With RT-PCR,the genetic expression of iNOS in myocardium was determined . Results: 1,In model group , after cardiogenic shock ,the plasma NO,NOS,iNOS,MDA levels increased;the activity of SOD decreased .2,Compared with model group, from 60 to 180 minutes , in shenfu group, the plasma NO,NOS, iNOS levels decreased significantly; the genetic expression of iNOS in myocardium was depressed ;in shenfu group From 30 to 180 minutes , the activity of SOD increased significantly ; the plasma MDA level significantly decreased; 3,The same changes in shenfu injection+dopamine group were observed.There were no significant difference between dopamine group and model group. Conclusions: These results suggested that excessive NO,NOS, iNOS,MDA and deficient SOD are responsible for the development of cardiogenic shock. The anti-shock mechanism of shenfu was probably that shenfu injection could inhibit the lipid peroxidation and increase the activity of SOD,depress the the genetic expression of iNOS.
