Objectives To study changes of distribution and content of connexin 43 andchanges of cardiac structure in microscope and electromicroscope in rat withacute carbon monoxide poisoning(ACOP), compareing the normal heart with theheart of rats with ACOP, trying to provide evidence of cytology biology forpathogenesis of myocardial damage in rat with ACOP, and academic base forclinic therapy of ACOP.Materials and Methods Thirty young male Wistar rats ,(3-5 months),weight:(300±25g)were randomly divided into ACOP group20 and control grouplO.(l)Making animal model: experimental rat model were ip injected with CO,170ml/kg wt, control group were ip injected with equal air. Sample of blood andcardiac tissue were taken after two hours, testing carboxyhe moglobin (HbCO)level in blood , doing electrocardiogram for rats.(2) Histochemistry staining:observe changes of myocardial structure by microscope.(3)Observe changes ofdistribution of connexin 43 through immunohistoche method, and changes ofcontent of connexin 43 through computer image analysis, (4) Observe changes ofmyocardial ultrastructure by transmission electro microscope. Rats of controlgroup and ACOP group were compared.Results1 Concentration of HbCO in blood increase obviously in rat with ACOP,ascomparing with control group, P<0. 05, there is difference.2, Heart rate of rat with ACOP is abnormal, there are various arrhythmia andmyocardial infarction in Electrocardiogram of rats with ACOP, as comparingwith control group, P<0. 01, there is remarkable difference.3 , Myocardial structure of experimental rats do not change in microscope by H-E...
