To quest for the evaluation system of Kidney-yang Deficiency rat Model whichaccord with TCM theoretically and experientially, including etiologicaldiagnoses, symptomatic diagnoses, laboratory inspect, therapy diagnoses, etc.For provide references to standardization work of TCM in animal model.Method:1.theoretical research:In this part, gave a systemic retrospect about the general development onanimal model of TCM; tidied up correlative literatures about Kidney-yangDeficiency, such as basic theory as well as methods of making such models;and summarized status quo about the study of animal model of Kidney-yangDeficiency.2.experimental research:2. 1 experiment â… : To select preferable method of making Kidney-yangDeficiency Model in male SD rat with HuIn this experiment, chose SPF male SD rat, 200-220g, used dosages accordingto the literature:190mg/kg, 280mg/kg, 375mg/kg, 450mg/kg, collected blood on7th, 12th, 17th, 22nd respectively, then drew dynamic curves according to thenumerical value on Cort, T, E2, T3, T4, cAMP, cGMP, etc. and compared withthe blank group in order to select preferable dosage and time consequently.2. 2 experiment â…¡: To observe the cardinal symptom affected by HuIn this experiment, made Kidney-yang Deficiency rat model which had established in the experiment I .observing the general response affected by Hu at the same time. All the rats were tested cold endurance capacity, provoked degree by snatching, run distance, autonomic activity, genito-function(male), etc. Results would be compared with the blank group. Through the symptoms observed supplement the objective index of experiment I , and modify the symptoms system of Kidney-yang Deficiency rat Model farther.2.3 experiment III: To observe the curative effect treated by Yougui Wan The rats were divided into three groups randomly: model group , Yougui Wan group and blank group. Model group and Yougui Wan group were took Hu 375mg/kg. w everyday, 17 days. Then the model group' s rats were detected Cort, T, E2, T3, T4, cAMP, cGMP, and organic pathology, while each Yougui Wan group' s rats were treated by Yougui Wan solution 4.5ml everyday. After 15 days Yougui Wan group' s rats and blank group' s rats were killed and detect objective index above too.2.4 experiment IV: To observe the stabilization of Hu ModelThe rats were divided into two groups randomly: recuperation group and blank group. Recuperation group were took Hu 375mg/kg. w everyday, while the blank group took saline. After 17 days, each group were feeding normally, 15days. Then all the rats were killed to detect Cort, T, E2, T3, T4, cAMP, cGMP, and organic pathology.Results:1. experiment I :Using Hu solution feeding rats, 375mg/kg. w per day for 17days, rats' Cort, T, E2, T3, T4, cAMP, cGMP, and organic pathology results accordant with pathological changes of Kidney-yang Deficiency. Establish the dosage and time of making Kidney-yang Deficiency rat model would be: 375mg/kg. w, 17days.2. experiment II :Making the Kidney-yang Deficiency rat model by the way established in experiment I . During the process, rats gradually losing weight, become emaciated, depilation, coat in abdomen wetted, and behave some symptom, such as languorous, sluggish and crouched. The average time of swimming in cold water, the ability of autonomic and burdened activity are lower than blank group, the genito-function(male) are greatly decreased too.3. experiment III: Yougui Wan group' s rats have recuperated after treated by Yougui Wan.
