Expression And Significance Of SR-A In Stroma Of Nasal NK/T Cell Lymphoma

IntroductionScavenger receptor A ( SR - A) is a kind of glycoprotein, and distributes on the surface of macrophages in kinds of tissues. SR - A gene ( MSR1) locates on 8p22, including 11 extrons. Different gene montafing can produce three subtypes , which are SR - A I , SR - A II and macrophage receptor with collage-nous structure ( MARCO). SR - A has six structure - function fields that can i-dentify different complements and plays an important role in pathophysiolicical processes, such as host defending, cell adhesive and apopsis. Additionally, some studies have discovered that SR - A is concerned with atherosclerosis, Alzheimer syndrome and endotoxin haemadrome.By far, study on the relationship between SR - A and tumor is not very much. Recently, someone detected that SR - A had close relation with prostate cancer. It expressed mainly on macrophages in stroma of prostate cancer and had many genetic changes. But the relation between SR - A and other tumors, such as nasal NK/T cell lymphoma, is not very clear. So we study the morphology and immunohistochemistry of nasal inflammation, nasal lymphoma in order to analyze the expression and significance of SR - A in nasal NK/T cell lymphoma.Materials and methods1. Materials95 cases were obtained from The First Affiliated Hospital of China Medical University from 1996 to 2000, and from 2002 to 2004, including 55 cases of na-sal NK/T cell lymphomas, 24 cases of B cell lymphomas and 16 cases of inflammations. In 55 cases of nasal NK/T cell lymphomas, male; 34, female. 21. The age was from 13 to 77, the mean age was 45.5. In 24 cases of B cell lymphomas , male15, female 9. The age was from 7 to 82, the mean age was 61. 9.2. ReagentsThe primary antibody, SR - A was provided by professor zhuwu of xiongben university in Japan. The ultro - sensitive S - P kit and DAB agent kit were brought from Fujian Maxin Biological Company.3. Methods(1 ) Slice preparing ? Every of paraffin - embedded specimens were chopped in 4 micron serises of slices for HE dyeing and immunohistochemistry study.(2) HE dyeing: Generally, slice was missed out wax to hydrate, stained by haematoxylin and eosin, then covered by general clarity gum.( 3 ) Immunohistochemistry ?. Slices were missed out wax to hydrate , by antigen repairing, dropped in non - antigen animal serum, incubated 30 minutes in 37 ^C. All the steps of SP immunohistochemistry were done by the introduction of S - P kit; joining up first and second antibodies, DAB, dyeing with haematoxylin again, slices were covered by general clarity gum.(4) Result determinant: SR - A protein was located in cytomembrane and cytoplasm, different brown granular reaction. SR - A positive cells excesslO% is regarded as masculine.ResultsSR - A expressed mainly in cytomembrane and cytoplasm, and SR - A positive cells were mainly distributed in macrophages of lymphoma stroma. The positive rate of SR - A was 92. 7% (51/55) in nasal NK/T cell lymphomas, 29. 2% (7/24) in nasal B cell lymphomas and 6. 25% (1/16) in nasal inflammation.
