Tissue Engineering is a modern subject. It includes a scaffold that provides an architecture on which seeded cells can be organized and developed into the desired organ or tissue prior to implant. The scaffold provides an initial biomechanical profile for the replacement tissue until the cells produce an adequate accellular matrix. Tissue engineered organs can help people to resolve the proplem of lack of organs for transplantation. Aims:To provide cells resource for the experimental study on urology tissue engineering. To establish the primary result of accellular umbilical veins as tissue matrix . to look forward to a hopeful alternative graft in clinical urology. Co-culture accellular umbilical veins with seeded cells . Methods:1 A bit of urethral mucous was taken from youngmale New Zealand hare and was digested by enzyme into single cell liquid. The cell was then cultured in a still condition. The medium was renewedregularly and the cells were subculeured and studied.2 umbilical veins were obtained from newborn and disposal in the base buffering use TRITON-X100 and PMSF . to wipe off the cell . holding the substance without immunity . the elastic and fibers were preserved. the vein accelular matrix were obtained. Co-culture accellular umbilical veins with the seeded cells. the condition of cells development and attach with the accellular matrix graft was observed with scanning electron microscopy and light microscopy examination. RESULTS:1 The urethral epithelium the cultured cells of young New Zealand hare can be cultured in vitro and is able to proliferate within a certain period. the morphological characteristics and immunohistochemistry of cells are accord with those of endothelial cells . 2 after three days. The accellular matrix was abtained. all cellular components were extracted and elastic and collagen fibers kept normal feature and structure . After 48h . The cells develop on the surface of graft were observed .some cells attach to matrix graft can be observed with the scanning electron microscopy . CONCLUSION:1 accellular matrix prepared from cell removing can be used as the scaffold for epithelial cells. 2 The method to provide umbilical veins accellular matrix is fitable .we can investigate more behavior in animal models.
